Objective To explore the effect and the mechanism enhancing fracture healing of zhuangjin xugu(ZJXG) decoction on the synthesis of type I and II collagen in callus of femur fracture rats.Methods The femur fracture model was generated by break off the femur at middle point with a line drama in 50 healthy adult male Wistar rats, and treated with ZJXG decoction for 28 days. The samples were collected at 7, 14, 21 and 28 days after treatment respectively. The fracture healing process was analyzed by gross anatomy and hematoxtlin-eosin(HE) staining in rats. The expression of type I and II collagen in callus was evaluated by immunohistochemical staining.Results In the sham operated group, the shape and structure of femur are normally and the expressions of type I and II collagen were weakly. In fracture model group, femur fracture-end was filled with blood tissue and the expressions of Type I and II collagen increased markedly. In control group, the fracture-end was filled with inflammatory cell infiltration and granular tissue at fracture 7 days, fibrous callus tissue at 14 days and cartilaginous callus tissue at 21 days, and the osseous callus until 28 days. In the treated group, the callus structure is better than that in control group. The expression of type I collagen decreased gradually from fracture 7 day to 28 day, while in treated group it was higher than that in control group during fracture 7-14 days. In control group, the expression of type II collagen increased successfully from fracture 7 day to 21 day, and reduced significantly at fracture 28 day, and in treated group it was higher than that in control group with no significant difference.Conclusion ZJXG decoction could promote the synthesis of type I collagen in callus during early stage(fibrous-cartilaginous callus stage) of femur fracture, which benefit to the formation of fibrous-cartilaginous callus.
