Objective:To study Zhuang Jin Xu Gu Decoction in promoting bone morphogenetic protein-7(BMP-7) and neuropeptideY (NPY) expression of femoral fracture callus, and discuss the fracture healing function of Zhuang Jin Xu Gu Decoction. Methods: Total of fifty adult and healthy male Wistar rats were included and prepared fracture model with saw cutting through the middle point of femur. The model group was treated by infusing the stomach therapy with Zhuang Jin Xu Gu Decoction. The fracture healing situation was observed by X-ray film, and the pathological change was detected by Hematoxylin-Eosin staining (HE). The expressions of BMP-7and NPY in fracture callus were determined by immunohistochemistry assay and the serum levels of BMP-7and NPY were detected by enzyme-link immunobsorbant assay (ELISA).Results: Seven days after treatment, the Wistar rat femur fracture swell disappeared; seven days after fracture, the broken ending was major fiber granulation tissue and was fibrous callus after fourteen days. While, seven days after treatment, fibrous callus was formed in broken ending, osteoid callus and bony callus was shaped after fourteen days. The expressions of BMP-7and NPY in fourteen days after fracture operation exceeded which in seven days remarkably, P<0.05. It was obviously higher in the model group than that in sham-operated group, P<0.05. The serum levels of BMP-7and NPY in treatment group were higher than those in model group, P<0.05.Conclusion:It is suggested that Zhuang Jin Xu Gu Decoction might promote the fracture healing by means of enhancing the expressions of BMP-7and NPY femoral fracture callus of rats.
