Objective The research object of this study was mongolian medicinal herb Juniperus rigida Leaves, to establish a quality control mode of HPLC fingerprint combined with QAMS(Quantitative Analysis method of Multi-components by Single marker) and an analysis method of GC fingerprint to provide the theory basis for the comprehensive control of the intrinsic quality of Juniperus rigida Leaves;and to establish a content determination method used Amentoflavone and Hinokiflavone contented in Juniperus rigida Leaves as the index of mongolian classical prescription “Jixiang Ankun pills”which content Juniperus rigida Leaves,to provide a scientific basis of Juniperus rigida Leaves for clinical application in mongolian medicine.Methods(1)HPLC method was adopted.Using Amentoflavone as an indicater,to establish fingerprint of 12 batches of different sources of Juniperus rigida Leaves;the RCF(relative correction factor) of Quercitrin 、 Podocarpusflavone A and Hinokiflavone were established,Then the RCF were used to calculate the content of Quercitrin、Podocarpusflavone A and Hinokiflavone,meanwhile the calculated value was compared with that of external standard method.(2)GC method was adopted.Using α-pinene as an indicater,to establish fingerprint of 10 batches of different sources of Juniperus rigida Leaves.(3)HPLC method was adopted to determine Amentoflavone and Hinokiflavone contained in Juniperus rigida Leaves used in Jixiang Ankun pills.Results(1)HPLC fingerprint common model of Juniperus rigida Leaves was established, 16 common peaks were marked, among which 5common peaks were identified: Skimmin、Quercitrin、Amentoflavone、Podocarpusflavone A、Hinokiflavone,the similarity of 12 batches of Juniperus rigida Leaves was more than 0.95;the RCF of Quercitrin、 Podocarpusflavone A and Hinokiflavone were 2.080、1.383 and 1.077 in turn, the RRT(relative retention time) were 0.559、1.254 and 1.303 in turn, The calculated value of QAMS was basically the same as the external standard method,and the relative error was not more than 1.08%.(2)GC fingerprint common model of Juniperus rigida Leaves was established, 13 common peaks were marked, the similarity of 10 batches of Juniperus rigida Leaves was not less than 0.90.(3)Method for content determination of Amentoflavone and Hinokiflavone in Jixiang Ankun pills was established,The linear range of Amentoflavone and Hinokiflavone were 0.0366 ~ 0.366μg, r=0.9999 and 0.0262 ~ 0.262μg, r=0.9999;with average recovery rates of 100.9%, RSD=1.22% and 99.7%, 1.81%(n=6).Conclusions The Quality control mode of HPLC fingerprint combined with QAMS for evaluation of the intrinsic quality of Juniperus rigida Leaves is comprehensive、accurate and exclusive,and GC fingerprint which mostly studies volatile ingredient in Juniperus rigida Leaves can be complemented each other with HPLC fingerprint information,and to better evaluate the quality of medicinal herb;Amentoflavone and Hinokiflavone in Jixiang Ankun pills are the main active components of Juniperus rigida Leaves,the content determination method is simple and easy,and provide reference for quantitative research of Juniperus rigida Leaves in Jixiang Ankun pills.
