Studies On The Monoterpenoid Indole Alkaloids In Gardneria Multiflora Makino And Uncaria Sinensis(Oliv.)Havil

Objectives: To study the monoterpenoid indole alkaloids in Gardneria multiflora Makino and Uncaria sinensis(Oliv.) Havil.Methods: Dried and powdered leaves of Gardneria multiflora Makino and the whole plant of Uncaria sinensis(Oliv.) Havil were extracted with 95% ethanol. After removal the solvent, the residue was extracted with petroleum ether and aqueous layer,then the aqueous layer was extracted with chloroform to obtain crude alkaloids. And the crude alkaloids were subjected to column chromatograghy on silica gel, MCI, Rp-18, HPLC, and Sephadex LH-20 to abeain pure compounds. The structures of compounds were elucidated by their physicochemical properties together with spectral data.Result: On the basis of their physical constant, spectroscopic data and modern analytic techniques, eleven monoterpenoid indole alkaloids and one non-alkaloids from the Gardneria multiflora Makino were elucidated, eleven alkaloids are gardmultimine A( YWX-1), gardmultimine B( YWX-2), gardmultimine C( YWX-3), gardmultimine D( YWX-12-2), gardmultimine E( YWX-7-1), gardmultimine F( YWX-7-2), gardmultimine G( YWX-7-3), 18-demethylgardneramine( YWX-4),demethocy-gardmultine( YWX-5), gardneramine( YWX-9), 19(E)-9,18-didemethoxygardneramine( YWX-10), and the non-alkaloids is Polystachyol( YWX-f2). Compounds 1 、 2 、 3 、 12-2 、 7-1 、 7-2 、 7-3 are seven new compounds, and among them 7-1 、 7-2 、 7-3 are three new Gardneria alkaloids containing with glucose units.Two monoterpenoid indole alkaloids from Uncaria sinensis(Oliv.) Havil were elucidated as Uncarine E( ZXN-1), Uncarine C( ZXN-3). Meanwhile, compound ZXN-1 and ZXN-3 were tested for cytotoxic aganist A549, HL60, K562 and BEL-7402 four human cancer cell lines in vitro.Conclusion: 11 monoterpenoid indole alkaloids isolated from Gardneria multiflora Makino, together with 2 monoterpenoid indole alkaloids isolated from Uncaria sinensis(Oliv.) Havil, were obtained by column chromatography and established by spectrum analysis. Among them seven compounds were new. Both compound ZXN-1 and ZXN-3 exhibited weak inhibitory against A549, HL60, K562 and BEL-7402 four human cancer cell lines in vitro.
