Association Of Serum Lipid And Gene Polymorphisms In Fujian Han Ischemic Stroke Patients

Objective: Investigating the distribution of PCSK9, LDLR, APOA5, APOB, APOE, SORT and other polymorphic loci in the ischemic stroke patients, and the relationship between the gene and blood lipid levels.Methods: a total of 435 patients with ischemic stroke were collected, and 200 patients with non cardiac and cerebral vascular diseases were collected as control group. Blood lipid related indexes and clinical data were collected. PCSK9, LDLR, APOA5, APOB, APOE, SORT and other polymorphic loci were detected by Sanger sequencing and mi LDR technology. At first, based on the detection results, testing Handy-Weinberg balance assessment, and analysis of differences in allele distribution of the study population and ethnic groups; and then, analysis of each genotype and allele frequency distribution between the case group and control group; again, analysis the blood lipid level between difference genotypes in case group; finally, the case group is divided into two groups, as the boundary of the LDL-C value 2.6mmol/L. Analysis the differences in the distribution of genotype and allele frequencies between the two groups.Results:(1) in 435 cases group and 200 cases in the control group gene polymorphism distribution analysis showed that: rs14158 A allele in the frequency of cases and control group respectively 39.9%, 23.2%, the difference is significant(=33.44, P = 0.000), with other sites in the difference between the case group and to contrast group does not have significant.(2) The analysis of genotype and blood lipid level in the case group:Relationship between the genotype and the level of TC: rs11583680 and rs7412, rs12740374, rs599839 carrying mutant gene of the population of the TC level is low, the difference is significant(P = 0.025, 0.013, 0.027, 0.002); rs7575840, rs688 carrying the mutated gene groups and rs2516839 heterozygous type high TC. The difference is significant(P = 0.037, 0.030, 0.025).The relationship between the genotype and the level of TG: rs662799, rs651821, rs2569556 mutant population level of TG is increased, the difference was significant(P = 0, 0, 0.035). The relationship between the genotype and HDL-C level: rs7412 mutant population of HDL-C is higher, the difference was significant(P = 0).The relationship between genotype and LDL-C level: rs7412, rs12740374 carrying mutant gene in the population of LDL-C lower, the difference is significant(P = 0.007 0.004); rs688 carrying mutations in genes of population and rs2516839 miscellaneous synthetic LDL-C higher. The difference is significant(P = 0.029 0.045).The relationship between the genotype and the level of APOA1 between rs2483205 and rs2495477 mutations in APOA1 is higher, the difference was significant(P = 0.016, 0.004)。The relationship between genotypes and APOB levels: site rs7575840 carrying population gene mutation can cause elevated levels of APOB protein, the difference is significant(P = 0.018); rs12740374 and rs7412 sites carrying population gene mutation can cause a decrease in the levels of APOB protein, the difference is significant(P = 0.004, 0.000) 。The relationship between the genotype and the level of APOA1/APOB: rs7412 with mutated APOA1/APOB gene in high value, the difference was significant(P = 0). Type LP(a) gene and the relationship between the levels of rs328 in people with mutated gene value is high LP(a), and the difference was significant(P = 0.023).(3) In the genotype frequency and LDL-C packet analysis tip: in high LDL-C group rs7412 carrying the mutation gene proportion of the population low, the difference is significant(=18.665, P = 0.000); site rs688, rs17671591 carrying gene population mutation frequency was high in the high LDL-C group, and the difference is statistically significant(P = 0.049 and 0.013). The genotype of other SNPs has no significant. Conclusion:(1) rs14158 allele of A in ischemic stroke with high frequency, but has no influence in the blood lipid level.(2) rs11583680 and rs7412、 rs12740374, rs599839 are related to the total cholesterol levels reduced. Rs688, rs2516839, rs7575840, to rs662799 are related to the elevated total cholesterol levels. Rs2569556, rs1260326, rs651821 are related to the high triglyceride levels; rs7412, rs12740374, rs688, rs2516839 are related to the decreased low density lipid protein cholesterol level. 3, rs2495477、rs7575840、rs12740374 and rs7412、rs328 are related to the levels of lipoprotein, but no significant with the ischemic stroke.(3) rs7412、 rs688、rs17671591 loci associated with LDL-C.
