Alangium chinense belongs to the Alangium genus of family Alangiaceae. It is warm, spicy and bitter. The root of this plant has historically been used as a traditional Chinese medicine for the treatment of rheumatoid arthritis, numbness, paralysis, heart failure, low back strain, bruises and other diseases. Pharmacology study has proved that alangium and its extracts have pharmacological effects, such as muscle relaxants, boost, anti-inflammatory, analgesic, anti-virus and anti-tumor.Objective: To explore the active ingredient and pharmacological effect of A.chinense, simultaneous determination method of 5 compounds from A.chinense was established by HPLC to study the differences in various organs of this plant, and chemical constituents were isolated and identified from the Et OAc extract of A.chinense.Methods: 1) The compounds were isolated and purified by various column chromatographies, and their structures were elucidated on the basis of chemical and spectroscopic analyses. 2) On the basis of the early chemical and pharmacological experiments, we determinated five active constituents simultaneously and compared the difference of the contents in different medicinal organs from A.chinense by the high performance liquid chromatography which was gradiently eluted with the mobile phase whose condition is organic phase(acetonitrile)- aqueous phase(0.2 g heptane and 3.5 g of sodium dihydrogen phosphate solution were mixed in 1 L flask).Results and Conclusions: 1) 9 compounds were obtained from the Et OAc fraction of A.chinense Et OH extract and identified as mansonone H(1),(1S, 4S)-7-hydroxycalamenene(2),(1R, 4S)-7-hydroxycalamenene(3), anabasine(4), gallincin(5),(6R,7E,9R)-9-hydroxy-4,7-megastigmadien-3-one-9-O-?-D-(6?-O-?-Dapi ofuranosyl)glucopyranoside(7),loganin acid(8),salicin(9),?-sitosterol(10).2)The results of the simultaneous determination using HPLC method showes that content of salicin, loganin acid, 6'-O-?-D–glucopyranosylhenryoside, anabasine and henryoside in the root of the A.chinense were 0.0621%, 0.1643%, 0.2405 %, 0.0284%, 0.0296%. The study also indicated that the contents of 5 compounds in different organs are quitely different.Lateral and fibrous root mainly contains loganin acid and anabasine, especially fibrous root contains large amounts of anabasine. There is only a little of 5 ingredients in the stem. Leave mainly contains salicin, 6'-O-?-D–glucopyranosylhenryoside and henryoside. Flower mainly contains salicin and henryoside. Fruit mainly contains 6'-O-?-D–glucopyranosylhenryoside and henryoside. The anabasine was not detected in in the leaves, flowers and fruits.Innovations : In this study, we determinated simultaneously the content of salicin, loganin acid, 6'-O-?-D–glucopyranosylhenryoside, anabasine and henryoside for the first time by HPLC and compared the diffence of content in different organs of A.chinense. This method is simple, feasible and stable, can be used for the determination and quality control of A.chinense, also can provide a new method and research basis for subsequent chemical analysis, establishment of quality standards, in-depth study of anti-inflammatory activity and development used as medicinal resource.
