Objective: To establish the oxygen-glucose deprivation and reperfusion model of BV2 cells. And to explore the effect of 14,15-EET on the inflammatory response of BV2 cells under OGD/R conditions.Methods: BV2 cells were cultured and identified. The condition of the OGD/R model of BV2 cells was confirmed by preliminary experiment. BV2 cells was randomly divided into three groups, namely, Sham group(normal cultivation group), Vehicle group(OGD/R intervention group), and 14,15-EET group(OGD/R and 14,15-EET intervention group). Under the treatment of 14,15-EET, the concentration of inflammatory factor in cell culture media of BV2 cells was detected by ELISA at different time points(0h, 3h, 6h, 12 h, 24h) after OGD1 h. The viability of BV2 cells was detected by MTT assay at different time points, and immunefluorescence staining of Ki67 was further applied to test the viability. At the same conditions, using wound healing assay and Transwell migration experiment, the ability to migrate of BV2 cells was observed.Results:BV2 cells was significantly impaired after OGD1 h, and its inflammatory secretion was significantly enhanced at OGD1h/R0 h, 3h, 6h, 12 h 24h. Compared with the Vehicle group, the 14,15-EET group had the less levels of inflammatory factor secretion, lower viability of survival and proliferation and weaker ability of migration. The above results are the most statistically significant at OGD1h/R12 h.Conclusion : The OGD/R model of BV2 cells was successfully established; and 14,15-EET can inhibit the inflammation and proliferation of BV2 cells induced by the injury of OGD reperfusion. It might be a possible target of anti-inflammatory therapy for clinical CNS disease.
