Objectives The aim of this study was to investigate the expression of Desmin in benign prostatic hyperplasia(BPH) and prostate cancer, and then analyze their correlation with the disease.. To further explore the involvement of Desmin in the development, invasion, and metastasis of prostate cancer. And explore the feasibility that Desmin as new biologic markers for diagnosis, prognosis and treatment for prostate cancer.Methods 1 Urological surgery or biopsy for prostate at the North China University of Science and Technology Affiliated Hospital,using random number table by transrectal patients of thirty cases diagnosed benign prostatic hyperplasia and thirty cases diagnosed prostate cancer,who met the inclusion criteria from January 2007 to October 2014.To obtain the corresponding biopsy from the hospital pathology department. 2 The expression of Desmin in 30 cases of prostate cancer and 30 cases of benign prostatic hyperplasia were detected by immunohistochemical SP method. The expression of Desmin in prostate tissue was detected. 3 According to the inclusion criteria from June 2015 to August, 4 cases of prostate cancer and benign prostatic hyperplasia were Frozen in liquid nitrogen who collected from the North China University of Science and Technology Affiliated Hospital.Western-Blot and Q-PCR method were used to validate the immunohistochemical results. 4 Analysis of the expression of Desmin in different pathological parameters of patients with prostate cancer.Results 1 In prostate cancer and BPH tissues, Desmin staining was detected predominantly in the Smooth muscle stroma of prostate,The positive stains were yellow or brown signals. Desmin expression was positive in 43.33%(13/30)of prostate cancer and in 73.33%(22/30)of BPH. The expression of Desmin was significant difference between benign prostatic hyperplasia and prostate cancer.2 Western Blot method show the expression levels of Desmin were significantly higher in BPH group than PCa group, the differences were all statistically significant. Q-PCR analysis the expression of Desmin m RNA were higher in BPH group than PCa group, the differences were all statistically significant.3 The expression of Desmin was significantly associated with T stage, Gleason score. Desmin expression was not significantly related to the age?N stage?PSA level?distant metastasis and Gleason score.Conclusions The expression Desmin were significantly higher in the benign prostatic hyperplasia than in prostate cancer. In the analysis of the clinical and pathological data of prostate cancer,The expression Desmin was significantly associated with T stage, Gleason score, Tip expression level and degree of differentiation of prostate cancer Desmin protein are correlated, that is related to the degree of malignancy of tumor cells. Desmin may influence the development of prostate cancer, and the diagnosis and evaluation of prostate cancer has important significance.
