The detection of drug resistance of mycobacterium tuberculosis is of great significance for prevention and treatment of tuberculosis(TB). In view of our national conditions, it is necessary to develop a quick, effective and cheap method for detection of multidrug-resistant mycobacterium tuberculosis.The plasmids carrying the drug-resistant mutation genes were designed for simulating different gene types of drug-resistant mycobacterium tuberculosis. The assay of multidrug-resistant mycobacterium tuberculosis were studied by combination of suspension array technology and allele specific primer extension reaction.The optimal reaction system of the assay was established. The developed assay in this study could simultaneously detect the multiple mutations, and there was no interference between each detected gene. The minimum detectable concentration was 1ng/m L; and repetitive experiments showed that the coefficient of variation(CV) of different gene mutation types ranged from 6.72% to 12.77%. This study has established a detection method of multidrug-resistant mycobacterium tuberculosis, which is characterized by high sensitivity,specificity, good reproducibility, rapid detection, low cost and high-throughput. The novel assay developed in this paper has great potential for clinical diagnosis of multidrug-resistant mycobacterium tuberculosis.
