Inflammatory bowel disease(IBD) is a group of diseases whose symptoms are chronic bowel inflammatory reaction. Abnormity of Immunity is considered to play an important role in the development of the disease. Cytokines are involved in the process of bowel immunity reaction and inflammatory procedure. In this study,we discuss the role of Interleukin-12 family cytokines in the process of IBD disease through detecting the levels of serum expression from 98 IBD patients and 24 patients with colon polyps respectively. It might shed light on the clinic treatment for IBD.ObjectiveTo investigate the serum levels of Interleukin-12 family cytokines:IL-12 、IL-23、IL-27、IL-35,expression in patients with inflammatory bowel disease and the clinical significance of them.MethodsAfter processing data quality,98 patients suffered from inflammatory bowel disease were enrolled. 61 patients were diagnosed as Crohn’s disease and 37 patients were diagnosed as ulcerative colitis. 24 patients with colon polyps were selected as the control group. Expression levels of serum IL-12, IL-23, IL-27 and IL-35 were detected using enzyme-linked immunosorbent assay(ELISA). Furthermore, we respectively analyzed the relationship between serum levels of expression IL-12, IL-23, IL-27, IL-35 expression and clinical features of inflammatory bowel disease.ResultsLevels of serum IL-12, IL-23,IL-27 from the ulcerative colitis group are significantly higher than those from the control group(P <0.001, P <0.001,P =0.023).Level of serum IL-35 from the ulcerative colitis group is lower than that from the control group(P <0.001). Levels of serum IL-12,L-23,IL-27 from the Crohn’s disease group are significantly higher than those from the control group(P<0.001,P<0.001,P=0.003). Level of serum IL-35 from the Crohn’s disease group is lower than that from the control group(P <0.001). Correlation analysis illustrates that levels of serum IL-12,IL-23,IL-35 are associated with the pathogenesis of IBD(OR=1.2501,P=0.0407;OR=1.9554,P=0.0101;OR=0.9204,P=0.0139). Levels of serum IL-12,IL-23,IL-35 are associated with incidence of ulcerative colitis(OR=1.2490,P=0.0411;OR=1.9413,P=0.0124; OR=0.9159,P=0.0147).Levels of serum IL-12,IL-27 and IL-35 are associated with the incidence of Crohn’s disease(OR=1.3964,P=0.0079;OR=1.0949,P=0.0469,OR=0.9408,P=0.0107). The level of serum IL-12 is positively related to the severity of Crohn’s disease and ulcerative colitis(P<0.001,P=0.0398). But the serum expression levels of other cytokines do not have obvious correlation with the severity of IBD.ConclusionLevels of serum IL-12 family cytokines are associated with the pathogenesis of IBD, whilst except for IL-12,other cytokines do not have obvious correlation with the severity of IBD.
