Objective To investigate the regulatory mechanism of bone marrow mesenchymal stem cell to the biological characteristics of KATO-III gastric cancer cells.Materials and methods Transwell cubicle was applied to build the co-cultured model in non-contact style.Then KATO-III cells were divided into two groups:co-cultured group and single cultured group.Each group was culture with or without stimulating factor SDF-1? or/and inhibitor AMD3100/LY294002.The stimulating effect of SDF-1? after 2 hours was observed with or without AMD3100/LY294002 pretreating KATO-III cells for 30 min.KATO-III cells cultured without SDF-1? acted as control and then each group was divided into different subgroups(Table 2)?All of the detecting target cells were KATO-III cells.The differences of cell proliferation and the resistance of anti-tumour drug(5-fluoropyrimidinedione, 5-FU and Cisplatin, CDDP) between groups were evaluated by Cell Counting Kit 8-assay(CCK-8).The invasion ability was detected by Transwell assay. The expression of CD133, CXCR4, apoptosis-related factors and EMT-related factors were detected by RT-PCR.Results Compared to single cultured group,BMSCs could regulate the stem cell makers expression of KATO-III cells and promote their proliferation ability(P<0.05).The growth rate of KATO-III cells in co-cultured group was significantly higher than that in single cultured group after treatment of 5-FU and CDDP(P<0.05).As compared to KATO-III cells in single cultured group, the number of infiltrating-membrane cells was significantly higher.The RT-PCR showed the relative m RNA expression levels of CD133,CXCR4, Bcl-2, N-cadherin, Snail in co-cultured group was significantly stronger than that in single cultured group,while the relative m RNA expression levels of Bax,E-cadherin in co-cultured group reduced(P<0.05).The proliferation, the invasion and the chemoresistance abilities of KATO-III cells treated with AMD3100 or LY294002 in co-cultured group declined as in single cultured group.While the relative m RNA expression levels of CD133, CXCR4, Bcl-2, N-cadherin, Snail in co-cultured group was significantly lower and the relative m RNA expression levels of Bax, E-cadherin in co-cultured group increased(P<0.05).Conclusions bone marrow mesenchymal stem cell could promote the CD133 expression of KATO-III cells and enhence their proliferation, invasion and chemoresistance abilities via SDF-1?/CXCR4-PI3K/Akt signal pathway.
