Part1: The analysis of rat liver cancer treat with HIF-1? by ultrasound-targeted microbubbles destructionObjective: To transfect rat liver cancer cells with HIF-1?sh RAN by ultrasound-targeted microbubbles destruction(UTMD)and explore its treatment effect for rat liver cancer.Methods: 36 wistar rats with liver cancer tumor, were randomly divided into two groups. The control group only received laparotomy without any treatment;while the intervention group received a complex treatment including UTMD,HIF- 1 ?sh RNA and microbubble(Sonovue). Contrast-enhanced ultrasonography images were acquired before the intervention and in day 3,day7,day14 after the intervention for both groups. Measurement of tumor size, tumor volume was also applied. 6 rats in each group were executed at different time points respectively, immediately after Contrast-enhanced ultrasonography imaging were acquired and the tumor tissue were taken out to,count microvessel density(MVD)after immunohistochemical staining with CD34.Result: 1. There were no significant differences between the two groups in tumor size before the intervention. The tumor tissue size was increased after intervention in two groups. Compared with time point before intervention, the tumor size was increasedsignificantly in the day 7 and day 14 after intervention in control group. Compared with day 7 after intervention, tumor size increased significantly in day 14 after intervention(P<0.05) in control group. However, the tumor size in intervention group was smaller compared with the control group in the same time points after intervention(P<0.05). 2. Tumor tissue microvessel density in intervention group is less than that in control group in the day 3,day 7,and day 14 after intervention. In control group, there were no significant differences in day3,day7,and day14 after intervention. In intervention group, it was reduced in day 14 after intervention, compared with that in day3 and days7 after intervention(P<0.01,P<0.05).Conclusion: UTMD technology can effectively transfect HIF-1? sh RNA to rat liver cancer cells and prevent tumor growth.Part2:Quantitative ass essment of changes in blood flow in rat live cancer by Contrast-enhanced ultrasondObjective: To discuss the value of the tissue blood flow by contrast- enhanced Flash-replenishment ultrasound in the evaluation of tumor anti-angiogenic gene therapy.Methods: 36 wistar rats with cancer tumor were randomly divided into two groups. Control group only received laparotomy without any treatment;Intervention group received a complex treatment including UTMD,HIF- 1 ?sh RNA and microbubble(Sonovue);.Contrast-Enhanced Flash-replenishment ultrasonography images before the intervention and in day 3,day 7,day 14 after the intervention were acquired for both groups. Data were stored in dicom format.to quantitative analyze the flow of blood in the tumor tissue using the Qlab workstation.Result: There were no significant differences between the two groups in tumor bloodflow before the intervention. The intervention group tumor blood was reduced compared with control group in the day3,day 7,and day 14 after intervention. There were no significant differences for tumor blood flow in control group in day3,day 7,and day14 after intervention(P>0.05). However, tumor blood flow in day14 after the intervention was less than that in day3 and day7 after the intervention(P<0.01,P<0.05). The tumor blood flow was correlated positively with MVD(r=0.73,P<0.01)Conclusion: Quantification of tumor blood flow by contrast- enhanced Flashreplenishment ultrasound can assess tumor anti-angiogenic gene therapy effectively.
