ObjectiveTo research the ratio CD4+CD25+CD127-/CD4+ Treg cells with psoriasis vulgaris,and the effects of different CG on the proliferation of Ha Ca T cells and the expression of IL-2 and IL-4 of Ha Ca T Cells,and study the correlation between pathogenesis of psoriasis and the ratio CD4+CD25+CD127-/CD4+ Treg cells and therapeutic mechanism of CG with psoriasis.MethodsPart one: Collecting patients information(sex, age,course of disease,PASI of skin lesion,Disease activity is advanced,stable and receding stage). The CD4+ CD25+ Treg of 30 patients with psoriasis vulgaris and 30 healthy volunteers were analysed by flow cytometry.and the correlation between the proportions of CD4+CD25+CD127-/ CD4+ Treg and PASI scores is analyzed.Part two: Ficoll density gradient centrifugation is used to separated PBMCs of 30 patients and 30 controls, The cultured Ha Ca T cells were intervened with the supernatants of PBMCs. The proliferation of Ha Ca T cells were evaluated by MTT assay. the expression of IL-2 and IL-4 was measured by ELISA. The correlation between the IL-2/IL-4 and PASI scores was analyzed.Part three: The Ha Ca T cells were cultured.The cultured Ha Ca T cells were intervened with CG which volume ratio is 0.05,0.1,0.2(low,medium and high concentration group)respectively. MTT assay was performed to evaluate the proliferation of Ha Ca T cells. The contents of IL-2 and IL-4 in Ha Ca T cells were examined by ELISA.Results1.The percentage of CD4~+CD25~+CD127~-/CD4~+ Treg was significantly decreased in advanced and stable psoriasis vulgaris than the control group(P < 0.05).The differences of between patient groups were statistically significant.There is strongnegative correlation between the percentage of CD4~+CD25~+ CD127~-/CD4~+ Treg and PASI scores(P<0.01).2.The proliferative levels of HaCa T cells stimulated by the lymphocyte supernatant of the patients were significantly up-regulated than those of the normal control.The secretion of IL-2 in patient’s PBMCs was significantly up-regulated in advanced and stable psoriasis vulgaris than negative control(P<0.05),The differences of IL-2 between patient groups were statistically significant. The levels of IL-4 of PBMCs in advanced group was obviously down-regulated than the negative group(P<0.05).3.The secretion IL-2 of supernatants with psoriasis were strongly and positively associated with PASI scores(P<0.01), moderate negative correlation exists between the secretion IL-4 of supernatants with psoriasis and PASI scores.4.CG between low, medium and high concentrations could obviously inhibited proliferation of Ha Ca T cells than control group,(P<0.01);CG of medium and high doses,the relative expression of IL-2 in Ha Ca T cells were obvious lower than the blank control respectively(P<0.01), The secretion of IL-4 in Ha Ca T cells was significantly higher than the blank group for treated with CG of high concentration(P<0.05),differences of IL-2 between drug groups were statistically significant(P<0.05).Conclusion1.The changes of CD4`+CD25~+CD127~-/CD4~+ Treg may be strongly negative correlated with the disease activity.2.The expression of PBMCs in psoriasis may promotes the proliferation of keratinocytes.3. The pathogenesis of psoriasis is explored by expression of IL-2 and IL-4,which are correlated with the disease activity.4.CG could suppress the proliferation of keratinocytes by regulating the secretion of cytokines IL-2 and IL-4.
