Objective To investigate the relationship between Helicobacter pylori(Hp) and extracellular regulated protein kinases(ERK1/2)in the pathogenesis of gastric carcinoma.Method The expression of Hp,ERK1/2,PKC~?andTRAF1 was comparatively determined by immunohistochemistry in 177 gastric specimens including 20 cases of chronic superficial gastritis(CSG),20 cases of intestinal metaplasia(IM),28 cases of dysplasia(Dys),36 cases of early gastric carcinoma,and 73 cases of advanced gastric carcinoma.Result The Hp demonstrated in the groups of Dys(39.29%) and IM(30.00%) were significantly higher than those in CSG(5.00%),early gastric carcinoma group( 2.78%) and advanced gastric carcinoma group(1.39%),respectively(P<0.05). There was no immunostaining of ERK1/2 to be shown in the groups of CSG,IM and Dys. The expression of ERK1/2 in the early(63.88%) and advanced gastric carcinoma group(68.49%) was detected. The rate of ERK1/2 expressed in undifferentiated adenocarcinoma group(82.50%) was obviously higher than that of well-differentiated adenocarcinoma group(51.52%,P < 0.05). It was shown that the immunoreactivation of PKC ~? found in the lymphocytes at the cancerous interstice only in the early and advanced gastric carcinoma group. PKC~?expressing in the lymphocytes of the well-differentiated carcinoma group(54.55%) was obviously higher than that of undifferentiated group(32.50%,P<0.05).It was shown that the immunoreactivation of TRAF1 found that there was a positive expression in the lymphocytes of all the samples,and there was no association between the factors.Conclusion It is indicated that the infective Hp in gastric mucosa induce the chronic gastritis,intestinal metaplasia,and dysplasia. The high-expression of ERK1/2 may be involved the progression and occurrence of gastric carcinoma. However,the significance of PKC~? and TRAF1 in gastric carcinoma are need further research.
