Objective:To observe acute toxicity of HUANGDANLAN granule in mice and chronic toxicity of HUANGDANLAN granule in rats. The inhibitory effect of HUANGDANLAN granule on Hca-F was observed by the experimental animal Hca-F liver cancer model. Determination of matrine and oxgmatrine in HUANGDANLAN granule by HPLC in order to establish the quality control method of HUANGDANLAN granule.Methods:1.The acute toxicity test of mice is about to be made of HUANGDANLAN granule suspension. Crude drug equivalent dose 0.4g/ml administered intragastricly to mice for one week. While recording the fluctuations of body weight in mice, as well as the change of spirit, loss of appetite, skin color, secretions, eye, respiratory, muscular exercise evaluation test in mice.2.Rat long-term toxicity test is about to be HUANGDANLAN granule into suspension, respectively, in low doses [0.4g/(kg·d)], medium dose [1.04 g/(kg·d)] and high dose [2.83 g g/(kg·d)] administered to rats.In accordance with standard once a day continuous intragastric administration a month to observe the rats hematologic indicators(including the clotting time, RBC, WBC, platelet count, WBC and hemoglobin content), urine routine, blood biochemical parameters(AST, ALT, Urea, Cr, CT) and the various organs(heart, liver, spleen, lung, kidney, adrenal gland, thyroid, testes or ovaries) and to carry out histopathological examination.3.HUANGDANLAN granule on experimental hepatocarcinoma Hca-F tumor bearing mice model effects by mouse hepatoma Hca-F mouse cells were injected into the modeling observed for HUANGDANLAN granule of tumor bearing mice with hepatocarcinoma Hca-F tumor inhibitory rate, effect on tumor bearing mice immune organ index and Hca-F in tumor bearing mice macrophage cell phagocytosis of chicken red blood cells were used as indicators, the discussion of for HUANGDANLAN granule on Hca-F inhibition effect and mechanism. Based on the level of determination of effects of HUANGDANLAN granule on immune function in tumor bearing mice serum IL-2 and VEGF expression in tumor bearing mice.4.Use the TIANHE C18(200mm×4.6, 5?m) column,its mobile phase was phosphate buffer solution(p H=3.0)- methanol(93:7).The flow velocity was 0.8m L·min-1 and the detective wavelength of Sodium Danshensu was 220 nm,and the column temperature is 35?.RESULTS:1.Acute toxicity test results in mice showed that the appearance of action in mice, reflex activity, water intake, large urinary conditions were normal.2. Compared with the control group, long-term toxicity tests in rats showed that participants spent three dose capsules against weight gain and general behavior of rats had no effect on activity; three hematology, urine and blood chemistry were measured no significant change. The major organs in rats treated group index and the control animals no significant difference. The histopathological examination showed the major organs in each group were non-toxic venereal reason to change.3.HUANGDANLAN granule on hepatocellular carcinoma Hca-F in mice model of experimental results show that the granule HUANGDANLAN granule can inhibit the growth of hepatocarcinoma Hca-F mice tumor, is more effective in the high dose, and HUANGDANLAN granule in a certain extent can improve the immune function of tumor bearing mice. The model index increase is most obvious in high doses. In the model, whole blood viscosity, plasma viscosity, hematocrit, and fibrinogen increased with high doses of more pronounced.4.It was showed that the matrine had a good linearity within the range of 2.5?g/ml~25.0?g/ml, the average recovery was 99.99%,and RSD=0.0577%(n=6). It was showed that the matrine oxide had a good linearity within the range of 2.0 ?g/ml~20.0 ?g/ml, the average recovery was 99.78%,and RSD=0.4432%(n=6).Conclusions:1.HUANGDANLAN granule is safe and non-toxic.2.HUANGDANLAN granule has good inhibition of mouse Hca-F hepatoma model of tumor growth and enhance the role of the immune function in tumor bearing mice. Huang Danlan granules to some extent, be able to improve immune function in tumor-bearing mice.3.The HPLC method is simple and accurate, and can be used as the quality control method of HUANGDANLAN granule.
