Objective :To investigate the correlation of insulin-like growth factor 2 binding protein 2(IGF2BP2) overexpression for glioma cell apoptosis and invasiveness.Methods :Construction GV146- IGF2BP2 eukaryotic expression vector, and using real-time PCR and Western Blot technique tested the expression level of IGF2BP2 gene in m RNA and protein situation of overexpression group, negative control group and blank control group of cell, to determine whether Construction is successful; Flow cytometry instrument detection IGF2BP2 expression effect on cell cycle; Transwell technology is used to inspect each cell invasion.Results:(1)Real-time PCR detection results: IGF2BP2 overexpressing cells of m RNA expression was significantly higher than the negative control group and blank control group, and no difference between the expression of negative control group and blank control group;(2)Western blot test results: Expression of IGF2BP2 protein in the overexpression group cell was higher than the negative control group and blank control group, Expression have no differs between the negative control group and blank control group;(3)Cell cycle flow cytometry results: The number of cell in G0 / G1 phase in overexpression group significantly more than the negative control group and blank control group, have no differs significantly between the negative control group and blank control group;(4)Transwell results: The number of cell which can pass through chambers in overexpression group significantly less than the negative control group and blank control group, have no differs significant between the the negative control group and blank control group.Conclusion :(1) GV146-IGF2BP2 eukaryotic expression vector was constructed successfully;(2)Expression of IGF2BP2 can arrest the cell cycle in G0 / G1 phase, and inhibit tumor cell proliferation;(3)Expression of IGF2BP2 can weaken the invasive of glioma cell.
