Objective To explore the effect and mechanism of anti-skin aging induced by acupoint injection with Stromal vascular fraction (SVF) combination with acupuncture Zusanli point.Methods SVF suspension was prepared by collagenase I solution and red cell lysate. Survival rate of SVF was determined by microscope after staining. Count of SVF was determined by the hemocytometer. A total of 24 rats aged 18 months were randomly divided into Group A, Group B, Group C and Group D. PBS was injected in Sanli acupoint of legs in Group A. Acupuncturing was given on Zusanli point of legs for ten days in Group B. SVF suspension was injected in Zusanli acupoint of legs in Group C. SVF suspension was injected besides acupuncturing was given in Zusanli acupoint of legs for ten days in Group D. Skin tissue was collected after rats were dislocated executed. Superoxyde dismutase (SOD), Hydroxyproline (HYP), Maleic dialdehyde (MDA) and Hyaluronic acid (HA) were determined respectively by xanthine oxidase method, dimethyamino-benzaldehyde method, thiobarbituric acid and enzyme-linked immunosorbent assay. The thickness of skin in each group was compared after paraffin sections of the skin tissue was stained by HE methods. Vascularization degree in each group was compared after paraffin sections of the skin tissue were immumohistochemically stained by ?-R Ag polyclonal antibody (1:100).Results (1) The contents of SOD, HYP and HA in Group B and Group C were significantly increased compared with those in Group A(P<0.05), and the content of MDA in Group B and Group C was significantly decreased compared with those in Group A(P<0.05). The contents of SOD, HYP and HA in Group G were significantly increased compared with those in Group B and Group C(P<0.05), and the content of MDA in Group G was significantly decreased compared with those in Group B and Group C(P<0.05). (2) The skin thickness and microvessel counts in Group B and Group C were significantly increased compared with those in Group A(P<0.05). The skin thickness and microvessel counts in Group D were significantly increased compared with those in Group B and Group C(P<0.05).Conclusion Acupoint injection with SVF combination with acupuncturing Zusanli point has favorable anti-skin aging through promoting the anti-oxygen ability by increasing content of SOD activity and hydroxyproline content,and lower malondialdehyde.Promote the skin blood vessels by SVF. And improve the thickness of the skin by increasing the hyaluronic acid and collagen content. Thus,combination of SVF and acupuncturing Zusanli point has better resistance to aging.
