High altitude polycythemia is one of the most frequently occurring and refractory diseases on the plateau.Its pathogenesis is complex and involves various systems of the human body,has so far failed to clarify.This experiment selected 42 male SD rats(Specific pathogen Free,240 to 270 g)randomLy assigned to control group(n=12)and model group(n=30).Model group combined with using Plateau environment simulation and cobalt chloride in SD rats to build a rat model of high altitude Polycythemia.And then,experimental Pathology and blood biochemistry methods,such as systematic study of high altitude Polycythemia in rats main organs and physiological and biochemical indexes of blood.The heart,liver,spleen,lung,renal tissue using Terminal deoxynucleotide transferase 2-dUTP-biotin Nick end labeling method for apoptotic cell detection,record the number of apoptotic cells.Aim is to enrich the model basis and clarify pathogenesis is a valuable thing.physiological and biochemical of blood:Compared with the control group,model Group rats' red blood cell count(RBC),hemoglobin(HGB),hematocrit(HCT)were significantly increased(P<0.01),in which hemoglobin concentration was greater than the 210g/L all of the Model gruop.Platelet count(PLT)and platelet hematocrit(PCT)decreased significantly(P<0.01).Model group:days winter ammonia acid amino transfer enzyme AST,Alanine aminotransferase ALT,Llactic dehydrogenase LDH activity obviously increased,and differences very significantly(P<0.01);Total bilirubin TB,combined bilirubin DB content very significantly sex increased(P<0.01);total protein TP,and globulin GLB content obviously increased,Albumin ALB content and Albumin/globulin A/G obviously reduced(P<0.01);Creatinine serum CRE,uric acid UA content obviously increased;Content of triglycerides TG was reduced(P<0.05)and TC of cholesterol,glucose GLU significantly reduce content(P<0.01).Pathological anatomy seen rats abdominal ascites with a small amount of blood,its' heart,liver,spleen,lung and kidney and other organizations the darker red.Also,the liver,spleen,lungs and kidneys showed obvious swelling.Visible under the light microscope:organizations of the model rats performed significantly congestion as well as hypoxemia.Bone marrow:hematopoietic cells are hyperactive,hematopoiesis function.Spleen:reticular fibers hyperplasia,evident megakaryocytes,manifestations of splenic xtramedullary hematopoiesis.Lungs:mainly two kinds of pathological changes severe terstitial pneumonia complicated by bronchitis:Around small blood vessels and pulmonary terstitial large number of inflammatory cell infiltration,bronchial mucosal epithelial cell amage and loss,significant pulmonary artery wall thickening.Diffuse pulmonary congestion nd hemorrhage:The structure of the lung disorder,serious congestion of blood capillary and ein,in the alveolar cavity filling of blood cell,diffuse status.Heart:mainly for the yocardial cells of myocardial gap widened,red dye.Liver:vacuolar degeneration of liver ells,liver sinusoids focal significant expansion.Kidney:renal Glomerular basement(?)embrane thickening,vacuolar degeneration of renal tubular epithelial cells,distal tubule and(?)he collecting duct luminal expansion cavity containing red dye substances.Ultrastructure of iisplay:Including cardiac myocytes,hepatocytes,kidney tubular epithelial cells,lymphocytes nd type I and II cells in the lungs:Severe swelling of mitochondria,disorganization broken dissolved,mitochondria vesiculation,and even as a whole disintegrated lisappear.Detection of cell apoptosis(TUNEL-POD)-positive cells:Model rats of spleen and ang cells apoptosis rate was significantly higher(P<0.01).Speculate that apoptosis(?)nechanisms may be involved in the HAPC spleen,lung tissue cells in the physiological egulation.Lliver,renal function and blood glucose and lipid metabolism disorders in model rats of(?)he High altitude Polycythemia.Hematopoietic function of bone marrow,spleen andr(?)xtamedullary hematopoiesis.Heart,liver,spleen,lung,and kidney tissue shows hypoxemia.
