Diabetes mellitus is a group of clinical syndromes of the disorder of glucose metabolism which is caused by a variety of factors. It has been drawing more and more attention due to the fact that it can result in so many complications; and it also has been called the third largest killer of human health following cardio-cerebrovascular diseases and malignant tumor because of its serious organ damage and high mortality. Recently, the research on DM is deepening gradually and more and more chemotherapy has arisen. Apart from insulin and its analogues, sulfonylureas, biguanides, thizolidinediones, a-glycosidase inhibitors, SGLT-2 inhibitors, GLP-1 receptor stimulants, and DPP-IV inhibitors are regarded as common clinical drugs. All above chemical drugs and protein drugs has shown advantages in the treatment of diabetes, such as high efficiency and significant effect, however, the use of them is often associated with the side effects such as hypoglycemia, weight loss, liver/kidney damage et al., which undoubtedly increases the pain of patients. What is worse, these drugs display no effect on DM complications; it is thus cannot improve the survival state of the patients.It is well known that Traditional Chinese Medicine (TCM) is a crystallization of the wisdom of our Chinese nation, and the application of modern technologies is largely accelerating its development. With the guidance of the basal theories and many practices of TCM, a growing number of scholars have turned over to extensively conduct research on Chinese herbal medicines. Up to now, hundreds of Chinese medicines for the treatment of DM patients and the improvement of their survival state have been developed based on the combined research of the modern pharmacological reagents and the traditional ancient medical skills. In addition, through further investigations on natural products, polysaccharides, saponins, flavonoids, alkaloids, and terpenoids have been demonstrated to play important and unique roles in the treatment of diabetes and its complications.Lamium barbatum var. Barbatum, a plant of Lamiaceae, also called Xuduan, can be found in three northeast provinces, Sichuan, Shaanxi, Gansu, Hunan, Hubei provinces and so on. Folk people usually use it to clear heat and remove toxin. DM has been generally regarded as fever by TCM, so Chinese herbs with the efficacy of heat-clearing and detoxicating are deemed to have good therapeutic activity on DM and its complications. In this study, the aerial parts of L. barbatum were extracted and purified through silica gel column chromatography and high performance liquid chromatography.20-Hydroecdysone, a flavonoid, three other compounds along with several fractions were obtained and their glucose consumption ability and hypoglycemic activity were evaluated on human liver L-02 cells. The results showed that the compound 20-hydroecdysone, the butanol extracts, the mixture of polysaccharides and flavonoids showed hypoglycemic activity at the concentration of 0.1 mg/mL in vitro, while flavonoid (compound 3) showed hypoglycemic activity at the concentration of 1 mg/mL.
