Objective:l.To study the effect of Xi-huang pills, Frankincense, Myrrh,and Ru-Mo pair on the growth of diverse human malignant tumor cell strains (MCF-7, MDA-MB-231, NCI-H460) in vitro and on the Matrix Metalloproteinases-9(MMP-9) of human breast cancer cell line MCF-7;2. To study the antitumous effect of Ru-Mo pair and its effect on receptor tyrosine kinases (RTKs) of human malignant tumor cell strains (SMMC7721, HL-60).Methods:1.The comparative study on theantitumous effects of Xi-huang pills, Frankincense, Myrrh, and Ru-Mo pair in vitro:?Adding the different concentration leachate of Xi-huang pill, Frankincense, Myrrh and Ru-Mo pair to 96-well cultumre plate, the proliferations of cells (MCF-7,MDA-MB-231, NCI-H460) were detected by MTT assay;?Adding the 4mg/ml concentration leachate of Xi-huang pill, Frankincense, Myrrh, and Ru-Mo pair to 96-well culture plate, the Matrix Metalloproteinases-9 of MCF-7 were detected by western blot assay.2.The study of Antitumous Effects on Ru-Mo pair:?Experment in vitro:Using leachate of Ru-Mo pair to treat diverse human malignant tumor cell strains(SMMC7721, HL-60)in vitro, proliferations of cells were detected by MTT assay, and the levels of RTKs by ELISA; ?Experment in vivo:The model of bearing H22 cancer mouse was established by transplantation, the mice were divided randomly into 5 groups:Ru-Mo high, medium and low dose group,cytoxangroup and control group. After continuous treatment for 10days,the tumors in each group were taken out and weighed for calculating inhibition ratio. Meanwhile, the level of RTKs in tumorswas detectedby ELISA.Results:1. The comparative study on the antitumous effects of Xi-huang pills, Frankincense, Myrrh, and Ru-Mo pair in vitro:?After the 48 hours'effection of the leachate of xi-huang pill, Frankincense, Myrrh, and Ru-Mo pair on the human malignant tumor cell strains(MCF-7, MDA-MB-231, NCI-H460), the growth of the tumor cell strains is inhibited(P<0.05; P<0.01),and with the rise of the medicine's concentration, its inhibition rate rise. ?After the 7 days'effection of th the Ieachate of xi-huang pill, Frankincense, Myrrh, and Ru-Mo pair on the MCF-7, The expression of MMP-9 on xi-huang pill group have obvious difference comparing with the control group(P<0.05), but the differences on the groups of Frankincense, Myrrh, and Ru-Mo pair were no significant (P>0.05).2.The study of Antitumous Effects on Ru-Mo pair:?Experment in vitro:At the concentration of 10mg/ml and 20mg/ml, the leachate of Ru-Mo pair show obvious depressant effects on cell multiplication of human malignant tumor cell strains(P<0.05; P?0.01), and obvious depressant effects of RTKs on SMMC7721 and HL-60 by Ru-Mo pair(P<0.05; P?0.01);?Experment in vivo:At the dosage of 1.08g/kg and 2.16g/kg,Ru-Mo pair shows obvious depressant effects of H22 mouse model (P<0.05; P?0.01), and obvious depressant effects of RTKs on H22 by Ru-Mo pair(P<0.05; P?0.01).Conclusion:Xi-huang pill, Frankincense, Myrrh, and Ru-Mo pair show obvious antitumous effects in vitro, and In breast cancer cell lines have a certain specificity.Ru-Mo pair is probbly the main herbs in the xi-huang Pills that could inhibit tumor cell, and the effect maybe correlate with depressing RTKs of malignant tumor cell.
