Fatigue is a comprehensive physiological process involving many physiological and biochemical factors.It’s a normal physiological phenomenon when the body’s mental or physical activity get to a certain stage.However,if the fatigue can not be relieved or eliminated for a long time,it may lead to a series of injuries to the body.So it becomes more and more concerned by the whole society that how to effectively alleviate the body fatigue.All along,the search for the efficient,safe,non-toxic anti-fatigue drugs aroused great interest in the field of medicine.In recent years,phytochemical resources are increasingly favored by the majority of scholars,which has wide application prospects because of its wide resources,small toxicity.As one of the China’s traditional Tibetan medicines,Rhodiola has a long history of medicine and it is a rare medicinal plant with a high medicinal and health value.Its active ingredient salidroside has anti-radiation,anti-fatigue and anti-aging and other important physiological effects,so it has a potential therapeutic effect to relieve physical fatigue.In additon,according to the theory of traditional Chinese medicine,several combinations of Chinese traditional medicines were prepared to create a multi-faceted,comprehensive anti-fatigue group through the appropriate compound.In this paper,we selected Rhodiola creulata from Tibet as the main raw material.The effective components of Rhodiola were extracted,and its crude extract was refined.The effects of the refining process on the loss of active ingredients of Rhodiola creulata were investigated.The ability to remove OH· and DPPH·,and the anti-fatigue activity of the extracts before and after purification were comparied and analyzed.Finally on the basis of single item recipe of Rhodiola,we choose Angelica,Schisandra,Cistanche,Chinese wolfberry,Polygonatum and other Chinese herbal medicine,after a proper mix,the anti-fatigue activities of each compound group and unilateral group were analyzed and compared,and the prescription of the best anti-fatigue effect was selected.The main findings are as follows:(1)Through the method of alcohol heat refluxing,the optimum extraction conditions of salidroside in Rhodiola creulata were determined by single factor and orthogonal test: 70% ethanol,solid-liquid ratio of 1: 20,75℃ for 3h.The yield of salidroside was 1.205%.(2)The flocculation effect of the two kinds of flocculants,gelatin and chitosan,was studied to refine the crude extract.The gelatin was selected as flocculant,and the ability to remove OH · and DPPH · of the refined extract was substantially equal to that before purification,both of which exhibited a strong ability to scavenge free radicals at low concentrations,and its ability to scavenge OH· is stronger than that of Vc(3)The male Kunming mice were used to investigate the anti-fatigue effect..They were randomly divided into blank control group,rhodiola crude extract of low dose group,middle dose group and high dose group,and salidroside control group,while the same grouping method was employed after the purification of crude extract.The indicators to be measured in this study include the exhaustive swimming time,serum BUN,blood LA,liver glycogen,etc.The results showed that rhodiola extract can greatly prolong the swimming time of mice.It can also reduce the contents of serum BUN and blood LA mice,and increase the content of liver glycogen.The trend of each index of the refined extract was similar to that of the crude extract,which was slightly lower than that of the crude extract,but there was no significant difference.The results showed that the refining process had no significant effect on the anti-fatigue activity with improving the taste,color,clarity of the Rhodiola extract.(4)According to the traditional Chinese medicine theory and the mechanism of fatigue,we selected the Angelica,Schisandra,Cistanche,Chinese wolfberry,Polygonatum among the traditional Chinese medicine that commonly used to relieve the fatigue though the ideas that mainly restore vital energy and promote blood circulation,in order to further improve the anti-fatigue activity.The Kunming male mice were randomly divided into six groups: quiet control group,exercise control group,formula groupⅠ,formula group Ⅱ,formula group Ⅲ,and single Rhodiola group.All mice were subjected to gavage administration for 30 days.The results showed that,compared with the exercise control group,all drug groups could obviously prolong the swimming time of mice,decrease the content of serum BUN,blood LA and MDA,increase the content of liver glycogen.Compared with single Rhodiola group,formula group Ⅲ exhibited a significant anti-fatigue effect.However,there was no signicant statistical difference between formula groupⅠ and single Rhodiola group.We further investigated the effects of each group on LDH,CK,SDH and ATPase activity in mice.Compared with the single group of Rhodiola,the effect of the group Ⅲ was significant(P <0.05).The results showed that other Chinese herbal medicine of formula group Ⅲ apart from Rhodiola can contribute to the anti-fatigue effect of Rhodiola,and it may have some directive significance for the development of novel anti-fatigue health products.
