Rhodiola is a kind of natural plant resource which can be both medicine and food ingredient. It has many functions such as anti-aging, radiation protection, anti-anoxia, anti-fatigue, tumour defending, improving immunocompetence and antivirus, etc. Salidroside is the primary functional component of Rhodiola and the method of extract salidroside from Rhodiola has been an important aspect in the R&D of Rhodiola medicines.In this paper, shaped circular ultrasonic extraction machine was used to extract salidroside from Rhodiola, and macroporous resin was used to purify salidroside. After elution, small molecule fraction including salidroside was concentrated by nano-filtration or rever-filtration, which then could be acted as materials in natural functional food, cosmetic and medicines. The main results were as follows:(1) The optimal parameters for the ultrasonic extraction of salidroside were determined by single factor and orthogonal experiments:Rhodiola powder was40mesh, concentration of ethanol was60%, ratio of solid to liquid was1:20, extraction temperature was40℃, ultrasonic power was600w (there is an interval every Is), extraction time was50min.(2) By examining the adsorption and desorption properties of nine macroporous resin, HPD600macroporous resin was chosen to be the optimal, the optimized parameters for the purification of salidroside were as follows:Rhodiola concentrated solution was diluted to10times as the sample, the saturation adsorption capacity of salidroside was7.4mg/ml resin, the eluent was40%ethanol, the pH of the sample solution was10and the pH of eluent was2.0.(3) After skimmed with petroleum ether and butanol extract,the content of salidroside reach to12.69%and40.92%, then through the HPD600macroporous resin,its purity is up to90.25%,which is improve the purity of salidroside further more. (4) Salidroside was determined by HPLC. The parameter was as follows:the measure wavelength was278nm, solvent was acetonitrile and water (1:9, v/v), the injection volume was10μl, column temperature was30℃, detection time is20mins. In above conditions, the detection time of salidroside was7.891min.
