Background and Objective:Endometriosis is defined as the presence and growth of endometrial glands and stroma out of the endometrium of the uterine cavity and the myometrium.Endometriosis is a benign disease,but it has some malignant biological behaviors such as proliferation,invasion,metastasis and recurrence.Now it is more inclined to think that endometriosis is a kind of tumor disease[1].There are many theories about the pathogenesis of endometriosis,and the most widely accepted theory is the theory of implantation,that is,endometrial fragments in countercurrent menstruation can grow in different places after adhesion,invasion and angiogenesis.The migration and invasion of ectopic endometrial cells is associated with epithelial mesenchymal transition(EMT).However,the specific regulatory mechanism is still unclear.PARP-1[poly(ADP-ribose)polymerase-1]is a nuclear protein enzyme widely exists in eukaryotic cells.It plays an important role in DNA repair,cell proliferation and differentiation,apoptosis,inflammation,and genomic stability[2].The expression of PARP-1 is increased in a variety of tumor tissues and PARP-1 can regulate the EMT process.However,the expression of PARP-1 in endometriosis and its relationship with EMT in endometriosis has not been reported at home and abroad.The purpose of this study was to investigate the role of PARP-1 in the pathogenesis of endometriosis by detecting the expressions of PARP-1 in eutopic or ectopic endometrium of women with endometriosis and normal endometrium without endometriosis.Study the relationship between PARP-1 and epithelial mesenchymal transition related markers of E-cadherin,vimentin,and transcription factor of Snail in eutopic and ectopic endometrium with endometriosis.Methods:Detectting expressions of PARP-1 and epithelial mesenchymal transition related markers E-cadherin,vimentin and Snail in eutopic and ectopic endometrium with endometriosis and normal endometrium by immunohistochemistry,real-time fluorescence quantitative PCR and Western blotting,respectively.The correlation between PARP-1,E-cadherin,vimentin and Snail expression levels were analyzed by Spearman correlation analysis.Results:1.The expressions of PARP-1,vimentin and Snail in eutopic and ectopic endometrium with endometriosis were significantly higher than that in the normal endometrium without endometriosis(all P<0.05).The expression of E-cadherin in eutopic and ectopic endometrium with endometriosis was significantly lower than that in the normal endometrium(all P<0.05).There were no significant difference in the expressions of PARP-1,E-cadherin,vimentin and Snail between the lesions of the eutopic and ectopic endometrium with endometriosis(all P>0.05).2.The different expressions of PARP-1,E-cadherin,vimentin and Snail in normal endometrium,eutopic endometrium and ectopic endometrium with endometriosis during proliferative phase and secretory phase were not statistically significant(all P>0.05).3.The expressions of PARP-1,E-cadherin,vimentin and Snail were correlated with the clinical stage of rASRM in endometriosis(P<0.05).4.In the lesions of the eutopic and ectopic endometrium with endometriosis,the expression level of PARP-1 was negatively related with the level of E-cadherin and the expression level of Snail,vimentin were positively related.Conclusions:The expressions of PARP-1 in eutopic and ectopic endometrium with endometriosis are higher and related with EMT markers(E-cadherin,vimentin,Snail).PARP-1 may be involved in the EMT process of endometriosis.
