Anoectochilus roxburghii is a precious perennial Chinese herbal medicine of the genus Orchidaceae.It is mainly cultivated in the market with many varieties and completed sources now.There is no effective quality evalutation to Anoectochilus roxburghii and it is always seriously admixtured and adulterated by others.Near-infrared spectroscopy has many advantages such as no sample destruction,rapid and environmental analysis.We collected the cultivated Anoectochilus roxburghii?Anoectochilus formosanus,Hongxia,and wild Goodyera,Ludisiain and we took the advantages of its Quantification and Qualitation to establish the models in order to detective its quality rapidly.The research could provide scientific basis for quality evaluation in the furture.The specific research contents were as follows:1.Based on NIR-PCA technology to build a cluster analysis model and distinguish Anoectochilus roxburghii from its close relatives.The qualitative discriminant model was evaluated by Q value,Cluster per Property,Spectra Residuals too big and Property Residuum.4400-4800?5400-6600 and 7800-10000cm-1 were selected as the modeling band,nle and dgl were used to pretreat spectral.The results show that the model of Anoectochilus roxburghii and its close relatives is predictive and the accurate is 98%.2.Based on NIR-PCA technology to build a cluster analysis model and identify the species of different cultivation patterm(3 months vitro seedling,4 months vitro seedling and 6 months of planting seedlings).The qualitative judgment model of Anoectochilus roxburghii in different cultivation stages was evaluated by Q value,Cluster per Property,Spectra Residuals too big and Property Residuum.Choosing 5000-10000cm-1 as the modeling band,nle and dgl were used to pretreat spectral.The results show that the model is predictive and accurate is 80%.3.Based on NIR-PCA technology to build a cluster analysis model and distinguish the sharp leaves,large round leaves,Hongxia,and Taiwan line of lotus from different cultivation methods(including 3 months vitro seedlings,4 months vitro seedling and 6 months of planting seedlings)with the Q value,Cluster per Property,Spectra Residuals too big and Property Residuum.The results show that the Q values of the four models were more than 0.9 and the accuracy was 100%,and the predictability of each model is good.4.The content of flavonoids in Anoectochilus roxburghii were detected by UPLC method,and the quantitative analysis model of Flavonoids from Fraxinus mongolicus was established based on NIR-PLS technique.4.1 Establishment of UPLC method for simultaneous detection of five flavonoids in 113 batches of Anoectochilus roxburghii:Narcissus,Rutin,Quercetin,Karethapine and Isorhamnetin.There was a good linear relationship in the five flavonoids between the concentration range(R2?0.9999).The average recoveries and RSD were between 97.06%-106.17%and 3.17%-6.24%.The content of Narcissus,Rutin,Quercetin,Kaempferol and Isorhamnetin in the tested samples were 217.17-2243.26ug.g-1,15.95-1414.26ug.g-1,11.87-267.07 ug.g-1,15.58-165.21 ug.g-1,13.54-86.4 ug.g-1.4.2 Based on the NIR-PLS technique and Sa3 + SNV spectral preprocessing method,the quantitative analysis of the composition of the Narcissus was established by using the whole spectrum as the analysis band.The SEC,SEP,Rc,Rv and Q values were 0.134,0.147,0.974,0.969 and 0.792,and the linearity and correlation coefficients of the calibration model are f(X)= 0.9478X + 0.0551,r = 0.9736.And predicted 10 batches of sample content,the average relative deviation is 6.09%,indicating that the model has a good predictive performance.4.3 Based on the NIR-PLS technique and SNV + MSC spectral preprocessing method,the quantitative analysis model of Rutin was established with full spectrum as the analysis band.The SEC,SEP,Re,Rv and Q values were 0.024,0.022,0.978,0.969,0.866,and the linearity and correlation coefficients of the calibration model were f(X)= 0.9569X + 0.0062,r =0.9782,and the average relative deviation was estimated at 4.50%.4.4 Based on NIR-PLS technique and MSC + Sa3 spectral pretreatment method,the quantitative analysis model of Quercetin was established by using full spectrum as analysis band.The SEC,SEP,Rc,Rv and Q values were 15.508,16.732,0.962,0.950 and 0.430.The linearity and correlation coefficients of the calibration model are f(X)= 0.9263X + 7.8829,r?0.9625,and the average relative deviation is 8.15%.
