This paper first examined the relationship between the contents of flavonoids which from different polar extracts(PEE:petroleum ether layer;EAE:ethyl acetate layer;NBE:n-butanol layer;WE:water layer)and organs and the antioxidant activity in order to determine its main active ingredients.Microwave assisted extraction of Anoectochilus roxburghii used response surface methodology to determine the optimum conditions and chose polyphenols as a response value.Finally,determined the antioxidant components of potential by analyzed the antioxidant activity in vivo and vitro,and qualitative analysis with UPLC-Q-TOF-MS/MS.A roxburghii was widely eaten with fresh.Therefore,after determined the antioxidative of pharmacological activity,the content of amino acids and mineral elements also should be determined and analyzed to evaluate the plant resource and to investigate the difference regularity.The result showed that the contents of flavonoids and polyphenols were leaf,whole grass,stem and root in the same extraction.The content of flavonoids was EAE>PEE>NBE>WE and the polyphenol content was NBE>EAE>WE>PEE in the same organ.The in vitro antioxidant experiment showed that the scavenging ability of leaves to free radical was significantly higher than that of other organs,the antioxidant capacity was NBE>EAE>WE>PEE.Polyphenol content of A.roxburghii had a positively related to antioxidant activity,that material basis of antioxidant activity may be phenolic.the optimum extraction conditions of microwave extraction were as follows extraction time:37.55 min;microwave power 320 W;ratio of material to liquid was 1:65.50;ethanol concentration was 83.90%;the yield of polyphenol was 1.31%.Antioxidant activity of the vitro showed that the maximum clearance rate of DPPH was 82.58%and the maximum clearance rate of ABTS+ was 97.63%.The vivo antioxidant had found the low dose group has good ability to scavenge free radical.Additionally,HPLC-Q-TOF-MS/MS was first used to determine 21 chemical composition in the extract.The results of this study provide a reference basis as a natural antioxidant.The principal component analysis result showed that the alanine,serine,glutamic acid,aspartic acid,threonine,isoleucine,phenylalanine,alanine,leucine were the characteristic amino acids and Fe,Zn,Mn,Ca,Cr,Mg were the characteristic elements of A.roxburghii,Cluster analysis showed that the distribution of contents had regional differences.The research results provide theoretical basis and direction for the development of a new type of foodstuff...
