Objective:To study the effect of eletroacupuncture at Changqiang(GV1)point on the 1 earning and memory function and expression of Neuroligin-3 protein in the hippocamp us region of the FMRI knockout mice(KO),and discuss its relevant mechanisms.Methods:(1)Breed FMR1 knockout mice by feeding one female and one male mouse i n the same cage.Then divide these newborn mice into three groups at random after ge ne identification by PCR.A total of thirty mice of FMR1 knockout mice were divide d into the eletroacupuncture "changqiang" group,the non-acupoint eletroacupuncture gro up and the model group,and each group contains ten mice.In the changqiang group,the eletroacupunture apparatus was set at continuous wave,2Hz,20 min,once a.day,six tim es a week for two weeks.The same eletroacupunture manipulation as changqiang group was used in non-acupoint group.The model group was fed under the same condition without any interventions.(2)Before(day 23)and after(day 41)the treatment,take theWater Maze Test to study learning and memory function of mice for each group.Do c raniotomy for each mouse after the treatment(day 46),then examine the expression of N euroligin-3 protein in the hippocampus region of the FMR1 knockout mice by immuo histochenmical staining.(3)Analyse the data by means of one-way analysis of variance.Results:(1)Water Maze Test:?the average escape latencies before treatments were:ch angqiang group(62.04±6.96)s?non-acupoint group(55.46±9.81)s?model group(57.83±17,72)s,there was no statistically significant difference between these three groups(P>0.05).;?the average escape latencies after treatments were:changqiang group(30.74±7.88)s?no n-acupoint group(40.82±8.05)s?model group(43.63±12.27)s,compared with the model group and the non-acupoint group,the changqiang group was decreased significantly(P<0.05);?the ratio of swimming time in platform quadrant to total swimming time bef ore treatments were:changqiang group(0.14±0.05)?non-acupoint group(0.17±0.08)?mode 1 group(0.16±0.07),here was no statistically significant difference between these three g roups(P>0.05).?the ratio of swimming time in platform quadrant to total swimming ti me after treatments were:changqiang group(0.31±0.06)?non-acupoint group(0.21±0.06)?model group(0..17±0.04),compared with the model group and the non-acupoint group,th e changqiang group was increased significantly(P<0.05).(2)Immunohistochemical:the gra y value of the positive expressions of Neuroligin-3 protein in the hippocampus region after treatment were:changqiang group(179.32±13.48)?non-acupoint group(196.69±11.80)?model group(197.615±7.99),compared with the model group and the non-acupointgroup,the changqiang group was increased significantly(P<0.05).Conclusion:Eletroacupunture at Changqiang(GV1)point can respectively improve learni ng and memory function of FMR1 knockout mice and the mechanism may be related to the improving of expression of Neuroligin-3 protein in the hippocampus region.
