Objective:To observe the effect of eletroacupuncture at changqiang(GV1) point on the expression of postsynaptic density-95 protein the hippocampus region of the FMR1 knockout mice.Methods:(1) First, select 2 cases sexual maturity FMR1 KO mices and among them, one female and one male mice were mated breeding. Then divide these newborn mices into three groups at random after gene identification by PCR. A total of thirty mices of fmr1 knockout mices were divided into the eletroacupuncture "changqiang" group, the non-acupoint eletroacupuncture group and the model group. In the changqiang group,the eletroacupunture apparatus were connected with continuous wave,2Hz,20 min, once a day, six times a week for two weeks. The same eletroacupunture manipulation as changqiang group was used in non-acupoint group where non-acupoint in the right rib was selected. The model group was reared under the same conditions without any intervention. (2)Before(23 days) and after the treatment, take the Water Maze Test for each group. Do craniotomy for brain next day after the treatment, then for immuohistochenmical staining and western blot methods on PSD-95 protein expression in the hippocampus region. (3)Analyses the data by means of one-way analysis of variance.Results:(1) Water Maze Test:①the average escape latencies before treatments were: changqiang group(59.61±7.46)s, non-acupoint group(56.76±8.96)s, model group (58.85±5.15)s, there was no statistically significant difference between these three groups(P> 0.05); ② the average escape latencies after treatments were:changqiang group(32.11±4.15)s, non-acupoint group(47.14±6.21)s, model group(49.50±6.87)s, compared with the model group and the non-acupoint group, the changqiang group was decreased significantly(P<0.05); ③the ratio of swimming time in platform quadrant to total swimming time before treatments were:changqiang group(0.16±0.03), non-acupoint group(0.18±0.04), model group(0.18±0.06), here was no statistically significant difference between these three groups(P>0.05);④the ratio of swimming time in platform quadrant to total swimming time after treatments were:changqiang group(0.40±0.11), non-acupoint group(0.32±0.08), model group(0.30±0.08), compared with the model group and the non-acupoint group, the changqiang group was increased significantly(P<0.05). (2) Immunohistochemical:the integral optical density of the positive expressions of PSD-95 protein in the hippocampus region after treatment were:changqiang group(0.21±0.02), non-acupoint group(0.18±0.03), model group(0.17±0.03), compared with the model group and the non-acupoint group, the changqiang group was increased significantly(P<0.05). (3)Western blot:the relative volume of PSD-95 protein in the hippocampus region after treatment were:changqiang group(1.48±0.14), non-acupoint group(1.02±0.16), model group(0.96±0.11), compared with the model group and the non-acupoint group, the changqiang group was increased significantly(P<0.05). Conclusion:Eletroacupunture at changqiang acupoint can respectively improve learning and memory ability in FMR1 knockout mice and the mechanism of the situation may be related to the improving of PSD-95 expression in the hippocampus region.
