| Objective Under the guidance of traditional Chinese medicine theory,the modern experimental technique was used to study the effect of Yiqi Huoxue Decoction on animal model of rheumatoid arthritis rabbits.We attempt to reveal the decoction pharmacological treatment and its mechanism from different angles,hoping to provide theoretical basis for clinical application.Methods 40 rabbits were randomly divided into normal group(NG),model group(MG),Yiqi Huoxue group(YHG)and Tripterysium Glycosides group(TPTG).The weight of AA rabbits,the perimeters of the hind toe were measured during different decades.After a following series of three-week therapy,the pathological changes of articular cartilage were observed;The levels of interleukin 17(IL-17)、23 in the serum and levels of IL-10、matrix metalloproteinase1(MMP-1)in the joint fluid of AA rabbits were measured by enzyme-linked immunosorbent assay(ELISA);The expression of Bax m RNA and Bcl-2 m RNA in synovial cells were detected by real-time PCR(polymerase chain reaction).Results 1.Effects of Yiqi Huoxue Decoction on general condition of AA rabbits: There was no significant difference in body weight and toe perimeters between all groups before modeling(P>0.05).After 3 weeks of therapy,the body weight of YHG was significantly higher than that of MG and TPTG(P<0.05),but showed no difference compared with the NG(P>0.05).Compared with the MG,the toe perimeters of YHG and TPTG was decreased significantly(P<0.05);compared with TPTG,YHG decreased notable,the difference was statistically significant(P<0.01);2.The results of ELISA showed that the levels of IL-17 and IL-23 in the serum of YHG and TPTG were significantly lower than those of MG(P<0.01);Compared with the TPTG,the leves of IL-17 and IL-23 was decreased,(P<0.05).The levels of IL-10 in the synovial fluid of YHQ and TPTG were increased than the MG(P<0.01),but there was no difference between YHG and TPTG(P>0.05).Compared with the model group,the levels of MMP-1 in YHG and TPTG were significantly decreased(P<0.01);Compared with TPTG,the levels of MMP-1 in YHG were decreased significantly(P<0.01).3.The HE staining showed that the articular chondrocytes of YHG and TPTG were relieved to some degree,and the chondrocytes were arranged in orderly rows.Compared with the MG,the articular chondrocyte destruction was alleviated to varying degrees,chondrocyte arrangement tends to normal,and the edema of tissues were also reduced.4.The PCR results showed that compared with the MG,the content of Bax m RNA in YHG was increased(P<0.01);Compared with the TPTG,there was no difference(P>0.05).The content of Bcl-2 m RNA in YHG and TPTG was significantly decreased compared with the MG(P<0.01);the content of YHG decreased statistically significant compared with the TPTG(P<0.05).Conclusions 1.Yiqihuoxue Fang has good anti-inflammatory effects on the RA model rabbit.2.Yiqi Huoxue Decoction can inhibit the release of inflammatory cytokines IL-17 and IL-23 in serum and control the inflammatory response.3.Yiqi Huoxue Decoction can reduce the level of MMP-1 in synovial fluid to ease the degradation of bone,and increase the level of inhibitory inflammatory factor IL-10 to improve the process of inflammation on the joint,which may prevent the RA pathological process.4.Yiqi Huoxue Decoction can regulate the content of Bcl-2/Bax,break the balance,and promote apoptosis of synovial cells,which contributed to the inhibitation of bone destruction. |
