In this paper,an immunochromatographic assay technology based cotton thread and utilized several nanomaterials was explored for rapid,sensitive and visual lung cancer biomarkers(squamous cell carcinoma antigen and human ferritin)detection.The main pionts of this thesis are summarized as follow:(1)Carbon nanotube reporter probe based cotton thread immunochromatographic assay methodIn this article,carbon nanotube(CNT)reporter probe based a cotton thread immunochromatographic assay technology was successfully prepared for visual and rapid detection of a lung cancer related biomarker,human ferritin antigen.The device can detect 50 ng/mL human ferritin antigen within 30 minutes,which improved the sensitivity approximately by 500 folds comparing with previous point-of-care test reports based on carbon nanotubes as reporter probe.(2)Double gold nanoparticle reporter probe based on a cotton thread immunosensor for electrochemical detection of lung cancer biomarker,squamous ell carcinoma antigenBy using two kinds of gold nanoparticle labels for improving detection sensitivity of SCCA,which greatly improved the sensitivity of the device comparing with employing traditionally a gold nanoparticle reporter probe base on cotton thread immunoassay technology.The assay avoids the multiple incubation and washing steps performed in most conventional protein analysis,shortened the analysis time and cost.Under optimal conditions,the linear range is 5?500 ng/mL.(3)Gold nanorods reporter probe based cotton thread immunochromatographic assay technologyIn this paper,a cotton thread immunoassay device combined with gold nanorods(GNRs)reporter probe was successfully developed for visual,rapid and sensitive detection human ferritin antigen(Human ferritin).Ferritin,a lung cancer protein biomarker,it is of great significance for early detection of lung cancer.Results indicated that the sensitivity of the cotton thread immunoassay device was improved nearly by 1000 folds comparing with traditional gold nanoparticles as reporter probe due to the unique optical properties of GNRs.Under optimal conditions,the cotton thread immunochromatographic assay technology can detect 5 ng/mL human ferritin antigen which meet the requirement for clinical diagnosis.
