In recent years,fingerprint of Chinese herbal medicine,which can be used to comprehensively analyze the quality of Chinese herbal medicine and to reflect the complexity and relevance of its effective components,is gradually accepted by the world.And it is a effective means to systematically evaluate the quality of Chinese herbal medicine.Building fingerprint of Chinese herbal medicine makes every medicine has its unique atlas,and those atlas can be used to solve problems existed in the study of Chinese herbal medicine.Thin layer chromatography(TLC)is the most common sort of chromatography,it is also the essential technology of fingerprint.TLC can separate multiple samples at the same time;can be apply to different components;can provide colorful images and multifaceted imformations of samples.But the traditional TLC also has lots of drawbacks,as a result,the resolution and reproducibility of the fingerprints are poor.High performation thin layer chromatography(HPTLC)made up for the drawbacks of classical TLC,and became more and more popular in modern labs.HPTLC uses automatic machines to replace the previous manual operations,which obviously improved the accuracy and reproducibility of analytical results.There is many reports of TLC fingerprint study of Chinese herbal medicine,and those fingerprints are clear and perfect.The number of Chinese herbal medicine is very huge.So,building the fingerprints of every Chinese herbal medicine is a process of accumulation over a long period.In this research,we gathered 10 different batches of several Chinese herbal medicine as the object of this research.Including Caodoukou,Hongdoukou,Yizhi,Gaoliangjiang,Jianghuang,Shannai,Bajiaohuixiang,Xiaohuixiang,Hujiao,Huajiao,Peilan,Shichangpu,Xinyi,Xixin and Fangfeng.We established the HPTLC fingerprints of those Chinese herbal medicine,and obteined the common patterns of HPTLC fingerprints of all these Chinese herbal medicine through CHROMAP 1.5 solution software.In addition we compared the similarity of different batches of all those Chinese herbal medicine,so as to evaluate the difference of chemical components among different batches of those Chinese herbal medicine.And the HPTLC fingerprint analysis we established in this study also can be used for rapid identification and quality control of all those Chinese herbal medicine.The main studies were summarized as followes:1.To collect 10 different batches of 15 kinds of Chinese herbal medicineWe collected 10 different batches of Caodoukou,Hongdoukou,Yizhi,Gaoliangjiang,Jianghuang,Shannai,Bajiaohuixiang,Xiaohuixiang,Hujiao,Huajiao,Peilan,Shichangpu,Xinyi,Xixin and Fangfeng,and extracted volatile oil components of those Chinese herbal medicine by the steam distillation method.2.To investigat the chromatography condition volatile oil components and other components of those Chinese herbal medicineThis study investigated the chromatography condition of all those Chinese herbal medicine comprehensively.Firstly we compared several different solvent systems to find the most suited one.After that we selected the temperature from 4℃、25℃、40℃,and selected the humidity from 18%、32%、47%、65%、72%、to establish a better chromatography condition of this study.In addition,we investigated the accuracy of the spotting machine,and also investigated the repeatability and stability of the test solution which we used in this study.Finally,We comfirmed ultima chromatography condition of all those Chinese herbal medicine.3.To establish the high performance thin layer chromatographic(HPTLC)fingerprints and the common pattern of those Chinese herbal medicine.The chromatography of this study were analyzed by utilizing CHROMAP 1.5 solution software.Through this software the fingerprint of those Chinese herbal medicine,with several characteristic peaks including reference substances,were set up.Furthermore,we created the common pattern of 10 different batches of those Chinese herbal medicine.Those specific HPTLC fingerprint that we established can be used for rapid identification and quality control of those Chinese herbal medicine.4.To analysis the similarity of 10 different batches of all those Chinese herbal medicineTo compare the data of different batches of those Chinese herbal medicine with the common pattern of them,we obtained the similarity data among them.Through similarity analysis we can evaluate the quality of those Chinese herbal medicine.The results revealed that Caodoukou,Yizhi,Gaoliangjiang,Jianghuang,Bajiaohuixiang,Xiaohuixiang,Peilan,Xinyi and Xixin showed higher similarity among different batches,while Hongdoukou,Shannai,Hujiao,Huajiao,Shichangpu and Fangfeng showed lower similarity among different batches.
