[Objective]This study aimed to investigate the effect of curcumin on the expression of miR-155 induced by inflammatory and the apoptosis of human trophoblast cells.[Methods]Human trophoblast cells (HTR-8/SVneo cells) were cultured and subsequently given different groups of treatments i.e. curcumin group (12.5,25,50 ?mol/L, added 0.5 h before incubation with cells), LPS group (100 ng/mL), and control group (blank medium). MiR-155 carrier was synthesized and the contents of miR-155 of HTR-8/SVneo cells were measured using real-time PCR method. The apoptosis of HTR-8/SVneo cells were tested by cell death detection ELISA. In addition, the contents of NF-?B (p65) of HTR-8/SVneo cells were investigated by western blotting.[Results]1?The expression of miR-155 was up-regulated by LPS treatment whereas the pre-treatment by curcumin could down-regulated the contents of miR-155, amongst which curcumin with 12.5 ?mol/L represented the most significant effect.2?Western blotting study showed that compared with control group all curcumin groups (12.5,25,50 ?mol/L) decreased the contents of NF-?B (p65).3?The result of cell apoptosis study indicated that compared with control group LPS treatment could induce the apoptosis while pre-treatment of curcumin (12.5,25,50?mol/L) showed different extents of inhibition.[Conclusions]The pre-treatment of curcumin could down-regulate the expression of NF-?B (p65) resulting in inhibiting the apoptosis of HTR-8/SVneo cells induced by up-regulation of expression of miR-155 by LPS treatment.
