| Objective:Human hepatocellular carcinoma Hep G2 cells was chosen as a model to investigate efficiency of curcuma longa extract and curcumin on apoptosis induction proliferation inhibition to Hep G2 cells and expression of inflammatory factors proteins.To investigate curcuma longa extract of anticoagulant,anti-inflammatory beffects and anti-inflammatory effect of curcumin on hepatocellular Hep G2 cell of liver cancer.Methods:Cultivate hepatocellular cell line Hep G2 with LPS,after 24 h,treat with curcuma longa extract and curcumin each. Proliferation inhibition was evaluated by MTT assay and calculate influence of half inhibit concentration(IC50).Cell morphological change and apoptosis were detected by FCM.Expression of inflammatory factors,including TNF-?,IL-1?,IL-6,i NOS,COX-2,PGE2,in Hep G2 cells was detected by Western blot.Results:(1)After 48 h treatment,proliferation of Hep G2 cells were inhibited with dose-dependent and curcuma longa extract IC50 value was 3.5mg/m L,curcumin IC50 value was 33?mol/L.(2)The G0/G1 phase of cells was down regulated and G2/M, S phase cells were up regulated.Under concentration of 1/2IC50,the apoptosis rate of the curcuma longa extract group is 31.42%,and the curcumin group is 25.28%.They compared with each other,there is no statistical significance(P>0.05).(3)The Western-Blot results showed,after 48 h effect,the curcuma longa extract and curcumin have a significant effect on anti-inflammatory by the expression of inflammatory factors TNF-??IL-1? and COX-2,and have no effect on IL-6?PGE2.Turmeric extract group have a downward effect on i NOS, suggesting that turmeric extract and other ingredients related to anticoagulation.Conclusion:1.curcumin is an important component of anti-hepatoma function in turmeric extract,play a major anti-hepatoma effect by anti-inflammatory pathway.2.Curcumin have a significant effect on anti-inflammatory by the expression of inflammatory factors TNF-??IL-1? and COX-2. |
