Tendon ruptured is a kind of soft tissue ruptured in sports or works. For the healing of tendon ruptured, surgery or physiotherapy is the main therapy.The ruptured tendon is not renewable so it can't repair completely itself when the tendon injured fearfully.Rupture tendon could be repaired by autologous transplant or allogenic transplant. The machanical strengh might restore less 40% as the normal through autologous transplantion. A lot of scar would exist as well.The allogenic tendon is low supply and it would become reject reation easily if allogenic transplanted in vivo.Using the biomaterial to replace the injured tendon will be the important way in the tendon repaired. Bacterial cellulose(BC) is sythesised by the bacteria with glucose in the comfortablee environment. BC has high purity quotient,water retention property and mechanical strengh.It's a natural nomameter cellulose with great biocompatibility and biodegradable.This research includ repairing the rat's ruptured tendon.Replace the injured tendon by the BC in the surgery and explore the tendon healing in original tissue engineering.The research of this thesis consist of 4 parts.The first part research is that gaining BC from the bacteria fermentation to be the biomaterial. Explore it's properties characterization in physical and chemistry.The study show that BC can be the replacement for the artificial tendon in physical and chemistry.The second part research validated that BC is not only less toxity fot the test cell but also make the test cell attach on it greatly. It prove that BC has the well biocompability.The third part research is replaced the rat's injured tendon by BC and observe in real time.During the replacement, the growth of the renew cells and the expression of the collagen show that the cells could attach on BC and even in it.The renew cells express the collage which consitute the renew tendon on BC.These renew cells and collagen provide the necessary condition for the tendon healing.The forth part research is the original tissue engineering. Making the bone marrow stem cells(BMSCs) in seed cells, BC in scaffold, to replace the rat's rupture tendon for the tissue engineering study.The results show that BC has high purity,crystallinity and water retention property.Its' biomechanics is stronger than the natural tendon so BC could satisfy animals' force.BC's cell toxicity is level I which is proved by the leaching liquor method.It shows that BC is nontoxic for the growth of cells.This accord with the require of the biomaterial.HE staining and immunohistochemistry explored the renew tendon cell could attach on BC and expressed collagen to healing injured tendon.The orginal tissue engineering research shows BC could be the biomaterial scaffold.The seed cells could grow on this scaffold and promote the repairing of healing.It also bring a new train of thought for the tendon repaired in the future.BC should have be uesd in the technology and theory of tissue engineering in the day to come.
