Objective:Based on the exsisting researches which find acupunture has effects on antianxi ety and regulating ANP-HPA axis, this essay further studies the effects of acup uncture on Oxytocin (OT) and Oxytocin Receptorin (OXTR) and corticosteron e (CORT) level of the terminal of hypothalamo-pituitary-adrenal axis (HPA ax is) of Chronic Stress Anxiety Rats. This article also discusses the working me-chanism of neuroendocrine of how acupuncture treating parts of anxiety disorde r, which may find new directions for studies on antianxiety by acupuncture.Methods:30 Clean grade and healthy male SD rats were randomly divided into 3 group s--blank, model and acupuncture,10 in each group. Chronic Unpredictable Stre ss (CUS) method was adopted to build animal model of anxiety. "Shenmen" a nd "neiguan" were selected to do hand needle therapy for acupuncture group.T he method of acupuncture is entwisting every 5 minutes, last for 30s, every ot her day for 1 time, around the turn, last for 30mins every time,started on the modeling day,which last for 15 days. The anxiety-like behavior was tested by using elevated plus maze.the contents of oxytocin and CORT in plasma was t ested by ELISA and the levels of oxytocin and OXTR in hypothalamus was te sted by immunohistochemicalResults:1.Results on ethology:Compared with the blank group,OT% of model group of elevated plus maze was decreased obviously (P< 0.05); Compared with model group, OT% of acupuncture group of elevated plus maze was increased obvio usly (P<0.05).It displayed that OE% of model group was decreased compare d to blank group, while OE% of acupuncture group was increased compared to model group, but there was no statistical difference between groups.lt was sug gested that the model animal were in anxiety state, which means model was w ell set and this anxiety state could be improved by acupuncture.2.Results on OT and OXTR in hypothalamus:Compared with the blank group, oxytocin level in hypothalamus of model group was decreased significantly (P < 0.01);Compared with model group, oxytocin level in hypothalamus of acupun cture group was increased significantly (P< 0.01),there was statistically signifi cance differences between groups.OXTR level of model group was decreased (P>0.05) and OXTR of acupuncture group was decreased compared with mod el group,while it showed no statistically significance differences (P?0.05);It dis played that acupunture could up-regulate the levels of OT in hypothalamus, but it didn't act on the OXTR level.3.Results on OT in plasma:Compared with the blank group, OT level of mod el group in plasma was decreased, and OT level in plasma of acupuncture gro upwas increased compared with model group.But there were no significance bet weeen groups(P> 0.05).4.Results on CORT in plasma:Compared with the blank group, levels of COR T of model group in plasma was increased (P<0.01);in contrast, levels of CO RT in plasma of acupuncture groupwas decreased compared to model group (P < 0.05); It's shown that CORT was rised by chronic stress.The results suggest ed that high CORT could be down-regulated with acupuncture.Conclusions:Acupunture could improve the anxiety status of model rats. This may relate to the functions of acupuncture:up-regulating the expression of OT in hypothala mus, down-regulating the CORT level in plasma and adjusting the function of HPA axis.
