At present,Parkinson's Disease(Parkinson 's diseases,PD)is one of the most popular popular neurodegenerative Disease,the incidence of Parkinson's Disease rank the second in neurodegenerative diseases,Parkinson's disease is more common in the population after the age of 65.Parkinson's disease is with complex clinical symptoms including motor symptoms such as tremor,slowness of movement,rigidity,and postural instability,and non-motor symptoms including personality disorder,mental disorders,sleep disorders and damaged sensory.Typical pathological feature of Parkinson's disease is the degenerative loss of dopamine neurons in substantia nigra,lower level of dopamine(DA).In mammals,dopamine plays an important role in motor coordination,emotion,memory and learning behavior.Now supplemntary of levo-dopa or Dopamine agonists is still an effective means in the treatment of Parkinson's disease.Many genes involved in synthesis,transport,secretion and metabolism of dopamine and its signal transduction pathways are conserved between flies and human.The enzyme tyrosine hydroxylase(TH)catalyzes the rate-limiting step in the biosynthesis of dopamine(DA)in dopamine neurons.In this research we measure the dynamic change of dopamine neurons in the physiological and biochemical level through the characterization of tyrosine hydroxylase's expression in dopamine neurons in genetic model of Parkinson's disease in drosophila.We establish genetic model of parkinson's disease in drosophila:TH-GAL4,UAS-a-SynA30P/TM2,TH-GAL4/Cyo;UAS-ParkinRNAi/TM6B,UAS-dLRRKI11915T/Cyo;TH-GAL4/TMB.We Used a series of monoclonal antibodies of tyrosine hydroxylase to immune dopamine neurons in the fly brain,statistics number of dopamine neurons in protocerebral posterior lateral 1.The experiment results show that compared with the control group in fruit flies:after 25 days,number of immune-positive dopamine neurons in the flies of Parkin RNAi and dLRRKI 1915T OE have reduced,reflecting the expression quantity of tyrosine hydroxylase in dopamine neurons in the flies of Parkin RNAi and dLRRKI1915T OE is lower.
