This experiment mainly includes three parts.Firstly,using the method of dual-frequency ultrasonic to extract the buckwheat hulls,and then using a series of chemical test methods(such as determination of total phenol content determination of flavonoid content,DPPH · radical scavenging activit,etc.)to determine the antioxidant capacity of buckwheat hulls extracts.At the same time,using high-performance liquid chromatography(HPLC)to analysis flavonoid polyphenols in buckwheat hull extracts.The results show that the content of total phenol and flavonoid are very high,reach 4.359mg/g,3.096mg/g respectively.And the content of rutin up to 3.055mg/g,the scavenging capacity for DPPH.and ABTS · reach 72.85%,46.86%respectively.Secondly,with the Wistar rats as the research object,using the method of vivo experiments to investigate the mechanism of buckwheat hulls on glucose lipid metabolism and oxidative stress in type 2 diabetic rats,diabetic rat models was established by injecting STZ.And the relevant mechanism of glucose lipid metabolism and oxidative stress related genes was discussed in diabetic rats.Results show that buckwheat hulls extracts intervention can reduce the content of MDA and raise the content of T-AOC,to reduce anti-oxidative stress injury caused by diabetes.At the same time,it can reduce the content of TC and TG to improve the ability of lipid metabolism,enhance the body's antioxidant capacity,and compared with the normal group and diabetes group,the intervention group significantly improved relative expression amount of mRNA on glucose lipid metabolism and oxidative stress-related gene.In the intervention group,glycogen synthesis genes(GS2,GYG1)upregulate,the gluconeogenesis pathway genes(PEPCK,G6PC1)downregulate,and the fat synthesis genes(ACC,FAS,SREBP1)were down-regulated.Through the study we found that buckwheat hulls extracts can effectively lower blood glucose in diabetic rats,improve the ability of lipid metabolism and increase resistance to oxidative stress,which greatly improves the utilization of buckwheat hulls.This mode is an efffective dietary intervention method for preventing the diabetes and improving symptoms of diabetes.Finally,the vitro experiment(cell experiment)further explores the buckwheat hulls extracts mechanism for lipid metabolism in C3H10Ti/2-Clone8,with adipogenic differentiation,analysis cells associated with lipid metabolism gene by the method of real-time PCR.The results showed that compared with adipogenic inducing cells,buckwheat hulls extracts intervention can significantly reduce the relative expression of mRNA on lipid metabolism related gene,which provides a theoretical basis for further improvement of glucose and lipid metabolism disorders in body.
