Study On The Protective Effect Of Salvia Tongluo Jiedu Decoction Combined With Bone Marrow Stem Cell Transplantation On Endothelial Cells In Rats With Acute Myocardial Infarction

Objective : Objective to study the protective effect and mechanism of Salvia miltiorrhiza Tongluo Jiedu decoction combined with bone marrow stem cell transplantation on endothelial cells in rats with acute myocardial infarction(AMI).Methods : The whole bone marrow adherence method combined with density gradient centrifugation was used to isolate and culture BMSCs.Myocardial infarction model was established by ligating the left anterior descending coronary artery(LAD).After the success,were randomly divided into SH(sham operation group),group AMI(AMI group),group C: group BMSCs(BMSCs transplantation group),group D:Chinese medicine group(Salvia Tongluo Jiedu Decoction group),group E: combined group(Salvia Tongluo Jiedu Decoction + BMSCs transplantation group,n = 10).BMSCs cell suspension was injected directly into the infarcted area of myocardial tissue.After 4 weeks,using enzyme-linked immunosorbent assay(ELISA)determination of serum nitric oxide in rats(NO),endothelin-1(ET-1)level,to observe the content of collagen Masson staining,immunohistochemistry observation of von Willebrand factor(vWF)protein expression,Western blot assay(WestNern Blotting)myocardial tissue thrombomodulin(TM)and endothelial cell protein C receptor(EPCR)protein expression level.Result: The results of this study show that,compared with the sham group,the level of ET-1 was significantly increased,NO decreased significantly,with differentdegrees of collagen fiber,vWF protein expression increased,the expression of EPCR and TM protein decreased,there was significant difference(P < 0.01);compared with the model group,stem cell transplantation group,Danshen Jiedu Tongluo Decoction group and Danshen Tongluo Jiedu decoction combined with ET-1transplantation group were significantly decreased,NO level increased significantly,reduced collagen fiber,vWF protein expression decreased,the expression of EPCR and TM protein increased,the difference was significant(P <0.01);compared with the stem cell transplantation group,Danshen Tongluo Jiedu Decoction group was significantly lower than the level of ET-1.The level of NO increased significantly,reduced collagen fiber,vWF protein expression decreased,the expression of EPCR and TM protein increased,the difference was significant(P <0.01);and the stem cell transplantation group,Dan Comparison of ginseng Tongluo Jiedu Decoction group,Danshen Tongluo Jiedu decoction combined with ET-1 transplantation group were significantly decreased,NO level increased significantly,reduced collagen fiber,vWF protein expression decreased,the expression of EPCR and TM protein increased,the difference was significant(P < 0.01).Research shows that Salvia Tongluo Jiedu Decoction can significantly reduce the level of ET-1,increase the level of NO and decrease collagen production,decrease the expression of vWF protein increased EPCR and protein expression of TM,endothelial protection,and combined with stem cell transplantation effect is stronger.Conclusion:1.Salvia Tongluo Jiedu Decoction can significantly improve endothelial cell protein C receptor in myocardial tissue(EPCR)and thrombomodulin(TM)expression,reduced von Willebrand factor(vWF)protein expression,decrease the serum level of ET-1,increase the level of NO,inhibition of myocardial tissue were collagen fiber hyperplasia,improve myocardial pathological morphology,inhibit thrombosis the formation and protection of vascular endothelial,and combined with stem cell transplantation effect is stronger.2.Salvia Tongluo Jiedu decoction combined with BMSCs transplantation cansignificantly reduce the serum ET-1 level of AMI rats,increase NO level,decrease collagen production,decrease the expression of vWF protein increased EPCR and TM protein expression,protecting vascular endothelium.
