Objective To study the effects of Invigorating Spleen Formula on rat neuron apoptosis situation and expression of PI3 K and AKT and Caspase3 and and Caspase8 after rat neuron is created a mode that lack of blood and oxygen.To explore the mechanism reduce the neuron apoptosis.Methods Separation and cultivate the neuron of newborn SD rat and assigned to six orifice plates.These plates divided into control group,hypoxia group,neural factor group,Invigorating Spleen group.Take these specimens into three gas incubator at 37? temperature.After cultivate 24 h,and cultivate 4h at the normal conditions.Use flow cytometry to detect neuron apoptosis;Use RT-q PCR and Western blot respectively to detect the m RNA and protein expression of PI3 K and AKT and Caspase3 and Caspase8.Results(1)Neuron apoptosis rates: Compared with control group,the hypoxia group neurons apoptosis rate are significantly increased(p<0.01);Compared with hypoxia group,the neurons apoptosis rate of neural factor group and Invigorating Spleen group are Significantly reduced(p<0.01);Compared with neural factor group,the neurons apoptosis rate of Invigorating Spleen group are Significantly reduced(p <0.01).(2)The expression of m RNA and protein of PI3 K,AKT,Caspase3 and Caspase8 : Compared with control group,the expression of PI3 K,AKT m RNA and protein of hypoxia group are significantly reduced(p<0.01),and the expression of Caspase3 and Caspase8 m RNA and protein are significantly increased(p<0.01);Compared with hypoxia group,the expression of PI3 K,AKT m RNA and protein of neural factor group and Invigorating Spleen group are significantly increased(p<0.01),and the expression of Caspase3 and Caspase8 m RNA and protein are significantly increased(p<0.01);Compared with neural factor group,the expression of PI3 K,AKT m RNA and protein of Invigorating Spleen group significantly increased(p < 0.01),and the expression of Caspase3 and Caspase8 m RNA and protein are significantly reduced(p<0.01).Conclusion The Invigorating Spleen formula can reduce the neuron apoptosis after cerebral ischemia,whose mechanism maybe related to impact the expression PI3K/AKT signal pathway and Caspase3 and Caspase8 that target gene lead to apoptosis.
