| Objective:By investigating the Effect of Formulae That Can TOnify Qi and Dissolve Stasis on VEGF ? TNF-? and ICAM-1 protein of Rats Model of Postpartum Endometritis,to explore the mechanism which Formulae That Can Tonify Qi and Dissolve Stasis intervened postpartum subinvolution of uterus caused by postpartum endometritis.Methods:40 pregnant SP rats were randomly divided into five groups.8 in each group.blank group,model group,Xinshenghua granule group and low,high dose of Formulae tonifying Qi and dissolving stasis group.Besides the blank one,from seventh days after conception,the rat model of qideficieecy and blood-stasis syndrome was established by using exhaustive swimming until delivery.First day after childbirth,endo-metritis model was established on rats by injecting escherichiacoli in vagina.With cold water,Xinshenghua granule,small,high dose of Formulae tonifying Qi and dissolving stasis by gavage in rats.All rats were executed after 8 days,and the count of VEGF?TNF-? and ICAM-1 protein were detected by the method of Immunohistochemistry.Result:(1)Under light microscope,in blank group,the inflammatory cell infiltration was unobserved and endometrial tissue was roughly normal.In the model group,a number of infiltrating inflammatory cells could be observed in endometrium,the degree of endometrium injury in Xinshenghua Keli group,low dose group and high dose was less than that in model group.Endometrium structure was normal in high dose group,interstitial part was dense and slightly edema,cell morphology was regular,evenly distributed,and had no blood ozzing and fluid ozzing,occasionally could see microvascular expansion and congestion.the degree of endometrium injury in Xinshenghua Keli group and low dose group was lessthan thatin model group.There was discontinuity of artial endocardium,and inflammationwas more severe than in model group.(2)The expression of VEGF protein in blank group rats uterine tissue increasedcompared with the model group(P < 0.01);Compared with the model group,after drug intervention,the expression of VEGF in uterine tissue in each treatment group increased,the differences were statistically significant(P < 0.01);There was nostatistically significant difference when compared Xinshenghua Keli group withlow dose group(P>0.05),but the expression of VEGF was significantly higher in high dose group than in Xinshenghua Keli group and low dose group,the differences were statistically significant(P< 0.01).(3)The expression of TNF-? protein was higher in model group than in blank group,the differences were statistically significant(P < 0.01);When compared with model group,the expression of TNF-?was decreased in Xinshenghua Keli group,low dose group and high dose group,the differences were significant(P < 0.05 or P < 0.01);the expression of TNF-?protein was significantly more in low dose group than in Xinshenghua Keli group(P < 0.01);There was no statistically significant difference when compared high dose group with Xinshenghua Keli group(P>0.05).(4)The expression of ICAM-1 protein was the lowest in blank group and was higher in model group(P < 0.01);When compared with model group,the expression of ICAM-1 was decreased in Xinshenghua Keli group,low dose group and hgh dose group(P < 0.01);There was no statistically significant differencewhen compared Xinshenghua Keli group with low dose group(P>0.05);but the expression of ICAM-1was significantly less in high dose group than in Xinshenghua Keli group and high dose group,the differences were statistically significant(P< 0.01).Conclusion:(1)The formulae that can tonify qi and dissolve stasis can control inflammatory reaction of the uterus of the rats with postpartum endometritis caused by after birth inflammation,alleviate tissues adhesion,promote endometrium angiogenesis and tissue restoration.(2)The expression of VEGF in the model group of postpartum endometritis was significantly lower than that in the blank group.After drug intervention,the expression of VEGF increased,Suggesting that low expression of VEGF may be an important factor affecting the uterine involution of model rats.The formulae that can tonify qi and dissolve stasis can promote involution of uterus by enhancing the expression level of VEGF.(3)After drug intervention,the expression of TNF-? decreased,suggesting that inflammatory reaction can increase the expression of TNF-?.The formulae that can tonify qi and dissolve stasis can reduce the expression of TNF-?,and reduce inflammation.(4)The expression of ICAM-1 was the lowest in the blank group and was higher in the model group.The expression of ICAM-1was lower in Xinshenghua Keli group,low dose group and high dose group than in the model group,indicating that abnormal high expression of ICAM-1 is the reason which leads to postpartum endometritis and affects the uterine involution of rats.The formulae that can tonify qi and dissolve stasis can reduce the expression of ICAM-1,alleviate tissues adhesion,reduce inflammation and promote involution of uterus. |
