Objective:This study was designed to determine the anaesthetic effect of propofol on alcohol dependence rats and alcohol withdrawal rats,and provide the experimental data for patients with alcohol dependence or withdrawal syndrome who need surgical treatment.Methods :(1)We compared the ED50 of propofol on alcohol dependence and normal SD ratsFifty male Sprague Dawley rats were randomly divided into two groups,alcohol dependence group and normal control group.The up-anddown sequential allocation technique was used to determine the median effective dose(ED50)and it's 95% confidence interval.The breath frequency,the onset time and the recovery time of the rats were observed.(2)We compared the ED50 of propofol on alcohol withdrawal and normal SD ratsFifty male Sprague Dawley rats were randomly divided into two groups,alcohol withdrawal group and normal control group,alcohol withdrawal rats were the alcohol dependence rats who cut off alcohol 24 hours.The up-and-down sequential allocation technique was used todetermine the median effective dose(ED50)and it's 95% confidence interval.The breath frequency,the onset time and the recovery time of the rats were observed.(3)We assessed the safety about 2ED50 of propofol on alcohol dependence-withdrawal rats by intravenous injectionAfter the treatment of the ED50 of propofol,rats of normal control group,alcohol dependence group and alcohol withdrawal group were raised one week to drug-eluting,then all the rat were received 2ED50 of propofol by intravenous injection.The breath frequency,the onset time,the recovery time,the activity complete recovery time and general situation changes of the rats were observed.Blood specimens and liver tissue of rats were taken for hepatic function biochemical detection and liver tissue HE staining.Results:(1)Twenty rats and sixteen rats were used in each group.The ED50 of propofol in alcohol dependence group was 9.56 mg/kg,95% confidence limit was 8.95~10.24mg/kg.The breath frequency was(92.3±2.72)bpm,the onset time was(9.3±0.87)s,and the recovery time was(513.9±15.48)s.The ED50 of propofol in normal group was 5.62 mg/kg,95% confidence limit was 5.02~6.30mg/kg.The breath frequency was(91.6±2.72)bpm,the onset time was(8.9±1.25)s,and the recovery time was(462.6±26.95)s.The ED50 of propofol of alcohol dependence group was increased thanthat of normal group.There was no difference about the onset time and breath frequency between alcohol dependence group and normal control group(P>0.05),while the recovery time of alcohol dependence group was longer than that of normal group(P<0.05).(2)Eighteen rats and seventeen rats were used in each group.The ED50 of propofol in alcohol withdrawal group was 4.09 mg/kg,95%confidence limit was 3.57~4.68mg/kg.The breath frequency was(94.8±3.19)bpm,the onset time was(9.8±1.64)s,and the recovery time was(571.2±42.23)s.The ED50 of propofol in normal group was 5.30mg/kg,95% confidence limit was 4.92~5.95mg/kg.The breath frequency was(92.5±1.94)bpm,the onset time was(9.9±1.27)s,and the recovery time was(479.6±88.97)s.The ED50 of propofol of alcohol withdrawal group was decreased than that of normal group.There was no difference about the onset time and breath frequency between alcohol withdrawal group and normal control group(P> 0.05),while the recovery time of alcohol withdrawal group was longer than that of normal group(P<0.05).(3)Rats were received 2ED50 of propofol,the breath frequency of alcohol dependence group was(95.8±4.09)bpm,the onset time was(8.6±1.14)s,the recovery time was(656.6±76.59)s,and the activity complete recovery time was(1257.2±92.35)s.The breath frequency of alcohol withdrawal group was(95.6±5.63)bpm,the onset time was(9.6±1.82)s,the recovery time was(528.2±42.14)s,and the activity completerecovery time was(1175.6±66.79)s.The breath frequency of normal group was(97.2±5.40)bpm,the onset time was(9.6±3.05)s,the recovery time was(500.4±39.40)s,and the activity complete recovery time was(1076.4±172.98)s.All rats reserved spontaneous breathing,and there was no obvious adverse reactions and 24 hours accidental death in alcohol dependence,alcohol withdrawal and normal group.There was no difference among groups in the onset time and breath frequency(P >0.05),the recovery time of alcohol dependence group and alcohol withdrawal group was longer than that of normal group(P < 0.05),the activity complete recovery time of alcohol dependence group was longer than that of normal group and alcohol withdrawal group(P<0.05),there was no difference among groups in the general situation,hepatic function biochemical test results and liver tissue pathology changes(P>0.05).Conclusion:Our study showed that the ED50 of propofol was increased in alcohol dependence rats,while the ED50 of propofol was decreased in alcohol withdrawal rats.It was safety and efficacy in all groups when rats were received 2ED50 of propofol.It may be safety to adjusting the dose of propofol according to the general condition of the patient with alcohol dependence or alcohol withdrawal.
