Introduction:The purpose of this research is to compare the impacts of Mineral Trioxide Aggregate(MTA,Dentsply,America)and iRoot BP Plus(Innovative Cerametic,Canada)on the proliferation and differentiation of human dental pulp stem cells(hDPSCs).Methods:Leach MTA and iRoot BP Plus with cell culture liquid and dilute the leaching liquids into different concentration ratios as 0,0.001,0.01,0.1,0.2 and 1;Test cell proliferation with CCK-8 kit after culturing hDPSCs 1 day,3 days and 7 days;Test alkaline phosphatase(AKP)expression after culturing hDPSCs 3 days and 7 days.Results:1.The number of cells increased significantly(P<0.05)with the increase of incubation time,but the MTA leaching solution wih concentration dilution ratio 1 has a significant cytotoxic effect on the growth of time.At the same time,iRoot BP Plus can promote the proliferation of hDPSCs better than the same concentration dilution ratio of MTA and has no cytotoxic effect.2.The expression of AKP significantly increases with time(P<0.05),but then it decreases when it comes to 1 concentration ratio of iRoot BP Plus.When treating hDPSCs with the same-concentration ratio of MTA and iRoot BP Plus after 3 days,the latter generally has a higher AKP expression except the 0.001 group,where MTA is higher than iRoot BP Plus(P<0.05);after incubated 7 days,hDPCSs express a higher AKP expression in the ratio of 0.001 and 0.01 group of iRoot BP Plus,and the latter has a significant difference(P<0.05)while in those higher concentration ratios------group 0.1,group 0.2 and group 1,the phenomenon adversely changes as MTA has a higher expression of AKP(P<0.05).Conclusions:High concentration of MTA has a cytotoxic effect on hDPSCs while the lower concentration exhibits a promotion of mineralization.iRoot BP Plus possesses a better biocompatibility and has no cytotoxic effect.Meanwhile,both materials suppress AKP expression to some extent and iRoot BP Plus inhibites AKP expression more when it comes to high concentration as time goes by.
