Objective: To analyze the Zedoary turmeric oil contents and its chemical constituents in four of different Curcuma processed products by GC-MS,and to determine the main components and compare the chemical constituents in Zedoary turmeric oil.To study the inhibitory effect of Zedoary turmeric oil on SW1463 cell line,and to explore its possible mechanism of action on SW1463 cell line.Methods:The volatile oil constituents of Curcuma phaeocaulis were extracted by steam distillation.And the main components contained in the volatile oil of Curcuma phaeocaulis were determined by GC-MS the analysis and calculation of the standard library.The Zedoary turmeric oil were prepared into a gradient of 40,80,120,160,200,240,and 280 mg·L-1.MTT colorimetric method to detect effects on the inhibition of cell proliferation that the different drug concentrations of zedoary turmeric oil intervene on rectal cancer SW1463 cells after 24,48 and 72 h.Calculate the inhibitory rate of Zedoary turmeric oil on cells and IC50.According to the drug IC50,Set the drug concentrationsinto 80,120,160,200mg·L-1and intervening thr cells for 24 h.The morphological changes of SW1463 cells were observed by inverted microscope and the Giemsa staining.The expression of Capase-3,Bax and Bcl-2 protein in rectal carcinoma cells was detected by Western blot.Results:78 chemical constituents were detected in volatile oils of four different processed Curcuma phaeocaulis,among which 58 constituents in the Fresh curcuma,53 constituents in the cut raw rhizoma zedoariae slices,52 constituents in the steamed Rhizoma zedoariae yinpian,51 constituents in the vinegar Rhizoma zedoariae.The main components of the volatile oils are monoterpenes and sesquiterpenes,but the contents and chemical constitutes of the volatile oil in different processedCurcuma phaeocaulis are quite different.MTT assay showed that Zedoary turmeric oil inhibited the proliferation of SW1463 cells ina time-dose-dependent manner.The 24 h,48 and 72 hhalf inhibition rate(IC50)was144.33,134.11 and 120.04 mg · L-1.Giemsal staining showed obvious morphological features of apoptosis.Western blot showed that 24 hours after the intervention,Compared with the negative control group,the expression of Caspase-3 and Bax was up-regulated(P <0.05,P <0.05),and the expression of Bcl-2 was down-regulated(P<0.05).Conclusion: 1.The main components of Zizhuo oil were monoterpene and sesquiterpenes,but the contentsand chemical constitutes of the volatile oil in different processed Curcuma phaeocaulis are quite different.2.Different concentrations of Guizhou Zedoary turmeric oil had a certain inhibitory effect on the proliferation of rectal cancer SW1463 cells.The inhibitory effect was enhanced with time and concentration,and it was time-dose-dependent.3.Zedoary oil can inhibit the proliferation and induce apoptosis of Rectal cancer SW1463 cells,and its mechanism may be related to up-regulation of Caspase-3 and Bax protein expression and to down-regulation of Bcl-2 protein expression.
