| Objective:To investigate the polymorphism of 15 STR(short tandem repeat)loci(CSF1PO、D7S820、D8S1179、D21SI1、D2S1338D3S1358、D13S317、D16S539、TH01、D18S51、D19S433、TPOX、vWA、D5S818、FGA)in Chinese Korean and Han nationality population in YanBian area,and evaluate its value of forensic medicine,for providing reference data for Individual identification,Paternity identification,Anthropology and other related fields.Methods:Sodium citrated blood specimens from 224 unrelated Chinese Korean nationality population in YanBian were collected,Sodium citrated blood specimens from 407 unrelated Chinese Han nationality population in YanBian were collected,The DNA were extracted from the specimens by method of Chelex-100 extraction.The 15 STR loci were amplified by using the AmpFISTR(?)Identifiler(?)Plus PCR Amplification Kit and screened by ABI Prism 310 Genetic Analyzer,and the solution were analyzed by software GeneScan Analysis3.7 and Genotyper3.7.The genetic parameters of each locus were calculated based on Modified-Powerstates statistical software.According to genetic frequency of selected 15 STR loci,we finally computed genetic distances among Chinese Korean and Han nationality population in YanBian area and 11 domestic ethnic groups.The phylogenetic analyses were made by Neighbour-Joining method of POPTREE2 software.Results:157 alleles and 415 genotypes were observed in 15 STR loci.The allele frequency and genotype frequency were ranged from 0.001-0.513 and 0.004-0.317,respectively.The heterozygosities(Ho)of the 15 STR loci were between 0.625 and 0.866 with average heterozygosity being 0.7732.The polymorphism information 0.800to 0.966,with the together discrimination power(TDP)being exceeding 0.999999999.The exclusion power(EP)were ranged from 0.322 to 0.727 with the cumulative exclusion power(CEP)being exceeding 0.99999729.158 alleles and 531 genotypes were observed in 15 STR loci.The allele frequency and genotype frequency were ranged from 0.001-0.546 and 0.002-0.309,respectively.The heterozygosities(Ho)of the 15 STR loci were between 0.604 and 0.853 with average heterozygosity being 0.7712.The polymorphism information content(PIC)was from 0.55 to 0.85.The discrimination powers(DP)were from 0.790to 0.966,with the together discrimination power(TDP)being exceeding 0.999999999.The exclusion power(EP)were ranged from 0.296 to 0.700 with the cumulative exclusion power(CEP)being exceeding 0.99999763.The genetic distance among Chinese Korean and Han nationality population in YanBian area and other ethnic groups which closed to it were smaller than that Chinese Korean and Han nationality population in YanBian area and other ethnic groups which far to it.The analysis of phylogenetic tree represent that the genetic relationship between Chinese Korean nationality population in YanBian area and the Korean nationality is the closest.Conclusion:15 STR loci(except TH01、D3S1358、TPOX)of Chinese Korean and Han nationality population in YanBian area are high polymorphism genetic markers.The loci are suitable for the Yanbian Korean and Han groups forensic identification.There are different degrees of difference between Chinese Korean nationality,Han nationality and other groups in Yanbian,therefore,it is necessary to set up the a group database. |
