ObjectiveTo summarize pathogenesis of postmenopausal osteoporosis from a theoretical point of view,and observe the changes of OPG/RANKL/RANK signaling pathway of ovariectomized rats with osteoporosis through experimental study.To observe the rapeutical effect and mechanism of Bushen Huatan formula on postmenopausal osteoporosis from the angle of adjusting bone metabolism of rats,and provide a scientific basis for the clinical application of compound recipt.Methods1 Theoretical InvestigationReviewing Literatures,studies show that OPG/RANKL/RANK signaling pathway is an important signaling pathway in bone metabolism,which can affect the pathogenesis of PMOP.Recalling the relevant literatures of the ancient and modern research PMOP,the cognition of the etiology and pathogenesis of PMOP mainly includes kidney deficiency,weakness of the spleen and stomach,insufficiency of liver blood,and blood stasis.2 Experimental StudyPostmenopausal osteoporosis rat model was established.Rats were randomly divided into model group,sham operation group,bushen group(0.51 mg/kg·b·w),bushen huatan group(0.94 mg/kg·b·w).5 weeks after operation,rats in treatment groups were intragastrically administrated for 8 weeks,rats in normal control group and sham operation group were administrated equivalent saline solution for 8 weeks.After rats were sacrificed,femur of left hindlimb was got,then levels of OPG and RANKL in femur were detected by western blot;blood in heart was draw and centrifuged,then contents of OPG and RANKL in serum were detected by ELISA assay.Statistical analysis: SPSS 13.0 statistical software were used for analysis of variance,all data were represented by mean ± standard deviation,P values of <0.05 were considered statistically significant.Conclusion1 Theoretical InvestigationOPG/RANKL/RANK signaling pathway is an important signaling pathway in bone metabolism,which can affect the incidence of PMOP from a theoretical point of view.Renal deficiency was regarded as key point to onset of osteoporosis in ancient and modern literature about pathogenetic factor and mechanism,therapeutical principle and method of osteoporosis.The topic raised hypothesis that abnormal lipid metabolism maybe related to osteoporosis,and put out new therapeutical method about osteoporosis;Bushen Huatan formula was clinical empirical formula of Professor Xiang Nan,it had better therapeutical effect on osteoporosis,and its effect on osteoporosis has been confirmed in animal experiments.And whether the prescription of bushen huatan can affect the OPG/RANKL/RANK signaling pathway remains to be further studied.2 Experimental ResearchCompared with sham group,the levels of OPG,OPG/RANKL in serum were significantly decreased and the levels of RANKL in serum were significantly increased in model group.Compared with model group,the levels of OPG,OPG/RANKL in serum were significantly increased and the levels of RANKL in serum were significantly decreased in bushen group and bushenhutan group.There was significantly difference between bushen group and bushenhutan group in above indices.Compared with sham group,the levels of OPG,OPG/RANKL in femur were significantly decreased and the levels of RANKL in femur were significantly increased in model group.Compared with model group,the levels of OPG,OPG/RANKL in femur were significantly increased and the levels of RANKL in femur were significantly decreased in bushen group and bushenhutan group.There was significantly difference between bushen group and bushenhutan group in above indices.
