Effect Of Strengthening The Spleen And Improving Intelligence On Promoting Value Differentiation Of Neural Stem Cells In MCAO Rats Based On Insulin/IGF-1 Signal Pathway

Based on the theory of "spleen govern transportation and transformation",this paper studies the therapeutic effect of spleen and puzzle on ischemic stroke from both theoretical and experimental perspectives,enriching the scientific connotation of "spleen master transport" theory,Chi-chi may promote the proliferation of neural stem cells through the energy metabolism,and continue to deepen the theory of splenomegaly,and provide new ideas for the clinical prevention and treatment of ischemic stroke.In the part of theoretical research,we first study the deep connotation of the theory of"spleen govern transportation and transformation":perfect the concept of spleen main transport,and put forward that the essence of spleen is the main content of the spleen and the main body.The internal operation of the spleen is the basic physiology of the main operation of the spleen,possession of the operation of the spleen is the main transport of deep gasification;Secondly,pointed out that "spleen store Ying and rich Ying" and "Ying cang yi zhu si" is the spleen The role of transport in the spleen-brain related mechanism,the key material related to the spleen and brain is the spleen transport of the valley,metaplasia of the camp gas;Finally,The relationship between spleen and brain was discussed from the perspective of the main application of spleen and energy metabolism.In the experimental study,MCAO model(middle cerebral artery occlusion model)was used as the carrier,and the blank group,model group,traditional Chinese medicine group,LY294002 group and LY294002 + Chinese medicine group were established.The survival time after the operation and points in each group was divided into the following three subgroups,respectively,1d,3d,7d.To evaluate the neurological deficits of the rats in each group for 2 hours to determine whether the model was successful.The cerebral infarction volume and serum insulin content were compared between the rats in each group.The expression levels of Brdu,BrdU/GFAP and Brdu/NeuN in hippocampus were detected by RT-PCR.The expression of Brdu,Brd/GFAP and Brdu/NeuN were detected by immunofluorescence.The results showed,after 3 days,the Chinese medicine group and LY294002 group,LY294002+ traditional Chinese medicine group compared Brdu immunofluorescence positive cells increased significantly;7 days after operation,LY294002 group Brdu positive cell number was significantly less than the model group,Chinese medicine group,LY294002+Chinese medicine group.It is suggested that Strengthening the spleen and improving intelligence can promote the proliferation of neural stem cells in ischemic side.The number of BrdU/GFAP double staining cells in the ischemic region of Chinese medicine group was significantly higher than that in group LY294002 and LY294002+Chinese medicine group,suggesting that Strengthening the spleen and improving intelligence granule can promote the differentiation of glial cells.New neurons did not see the Brdu/NeuN experiments,considering the possibility that the experimental cycle is too short,the neural stem cells do not differentiate into neurons.The Chinese medicine group rats hippocampus AKT,PI3K gene expression was higher than that in LY294002 group,Chinese medicine group the expression of PTEN was significantly lower than that of LY294002 group,suggesting that Strengthening the spleen and improving intelligence granules by promoting PI3K/AKT activation,expression of PTEN control,has protective effect on the brain.
