ObjectiveAnimal experiment To explore the effect of YIZHINING to the working memory of Spontaneously Hyperactivity Rat(SHR)-----The animal model of Attention Deficit Hyperactivity Disorder.Clinical research To evaluate whether Yizhining could improve the working memory of patients of ADHD of deficiency of kidney and hyperactivity of liver-yang,through n-back software.MethodAnimal experiment 50 SHR were randomly divided into five groups,Yizhining high,medium and low dose group,Methylphenidate Hydrochloride group(MH),and normal saline group,each group 10 rats.Five groups were treated by YIZHINING decoction(14.4g/kg,7.2g/kg,3.6g/kg)?Ritalin(0.0015/kg)and normal saline respectively.The gastric volume was equal(12.5ml/kg),once a day.After 4 weeks,Morris Water Maze test was useded to examine the working memory of each group.Clinical research 40 patients of ADHD of deficiency of kidney and hyperactivity of liver-yang were divided into two groups,Yizhining group and Xiao-er Zhi-li syrup group,20 cases in each group,working memory was tested using n-back software before the treatment,after 8 weeks,examine working memory again.ResultAnimal experiment 1)navigation experiment The escape latency of normal saline group undulated.more longer was the training duration,more shorter was the escape latency of the treatment group.At the fifth day,significant.difference surfaced between them(p<0.05),and Yizhining high dose group and methylphenidate group stood out(p<0.05),but the difference between these two groups was not significant(p>0.05).2)Space exploration experiment Compared with normal saline group,Yizhining high dose group swan longest in the target quadrant,Yizhining high dose group and methylphenidate group stayed more.time in the target quadrant,and crossed platform more times,and the difference was notable(p<0.05),but.the Yizhining high dose group and methylphenidate group showed no significant difference(p>0.05).Clinical research The deference of correct response times and correct click average time of the two groups were not significant before and after the treatment,while the correct response times of 40 cases remarkably increased after 8 weeks' treatment and the correct click average time outstandingly decreased compared with prior treatment.ConclusionAnimal experiment High dose of Yizhining can improve the working memory of SHR rats,and the effect is comparable to that of methylphenidate.Clinical research Yizhining could effectively improve the working memory of ADHD of deficiency of kidney and hyperactivity of liver-yang,and the difference was compared with Xiao-er Zhi-li syrup was not significant.
