| Objective:Recently,the research of regulatory T cell in the immunologic mechanism have been popular in caner and autoimmune diseases with the development of tumor microenvironment and tumor immune regulation.Thymic stromal lymphopoietin can promotes the differentiation of Tregs through dendritic cell.In this study,we mainly focus on the expression of TSLP and FOXP3 and the relationship between TSLP and FOXP3 in Hashimoto’s thyroiditis(HT),papillary thyroid carcinoma(PTC)and PTC coexisting with HT.Moreover,we analyze the relationship between TSLP and FOXP3 and the clinic pathological significance of TSLP and FOPXP3,furthermore,study the immunologic mechanism of them in PTC coexisting with HT.It is designed to provide new ideas for clinical diagnosis and treatment in thyroid disease.Method:There are 189 thyroid cases from the thyroid and breast surgical department of Subei Hospital between 2012 to 2015,including 25 cases of nodular goiter,54 cases of Hashimoto’s thyroiditis,64 cases of papillary thyroid carcinoma,46 cases of PTC coexisting with HT.The selection criteria eliminated that the patients have accepted preoperative treatments of thyroid gland,including radioiodine therapy.The expressions of TSLP and FOXP3 were detected by immunohistoehemistry SP staining in 189 samples of thyroid tissue.Result:The positive rate of TSLP and FOXP3 expression was respectively 8%and 16%in nodular goiter,83.3%and 70.3%in Hashimoto’s thyroiditis,82.8%and 60.9%in papillary thyroid carcinoma,95.6%and 89.1%in PTC coexisting with HT.The positive rate of TSLP and FOXP3 expression in Hashimoto’s thyroiditis group(χ2=40.243,p=0.000),(χ2=20.288,p=0.000),papillary thyroid carcinoma group(χ2=42.620,p=0.000),(χ2=13.538,p=0.000)and PTC coexisting.with HT group(χ2=54.549,p=0.000),(χ2=37.322,p=0.000)was significantly increasing,compared with nodular goiter group,the difference was statistically significant.Furthermore,the positive rate of TSLP and FOXP3 expression in PTC coexisting with HT group was higher than that in Hashimoto’s thyroiditis group(χ2=3.850,p=0.049),(χ2=5.269,p=0.022)and that in papillary thyroid carcinoma group(χ2=4.233,p=0.039),(χ2=4.233,p==0.001),the difference was statistically significant.According to clinical data analysis,the positive rate of TSLP expression in PTC coexisting with HT had relationship with lymphatic metastasis(χ2=3.920,p=0.048),while had no relationship with gender,age,tumor size and TNM stage(χ2=1.012,p=0.314;χ2=2.971,p=0.085;χ2=0.915,p=0.339;χ2=6.509,p=0.011).The positive rate of FOXP3 expression in PTC coexisting with HT had relationship with lymphatic metastasis(χ2=10.518,p=0.001)and TN-M stage(χ2=1.012,p=0.314),while had no relationship with gender,age and tumor size(χ2=0.139,p=0.709;χ2=0.809,p=0.368;χ2=2.374,p=0.123).The difference was statistically significant.The expression of TSLP was positively correlated with the expression of FOXP3 in PTC coexisting with HT(r=0.610,p=0.000).Conclusion:The expression of TSLP and FOXP3 had relationship with lymphatic metastasis,the expression of FOXP3 was related to TNM stage,while they had no relationship with gender,age,tumor size.The expression of TSLP was positively correlated with FOXP3,they play a synergistic role in PTC coexisting with HT,may be involved in the immune escape and immune suppression of in PTC coexisting with HT and had an impact on the occurrence,development and metastasis of PTC coexisting with HT disease. |
