Effects Of Yun Spleen Rou Liver Recipe On Brain-gut Peptide In Constipation-predominant Irritable Bowel Syndrome Rats

Purpose:Yun Spleen Rou Liver Recipe is the mentor's experience side,In this study,we established a rat model of constipation-induced irritable bowel syndrome,to observe the effects of Yun Spleen Rou Liver Recipe on the serum 5-HT?VIP?SP,colon tissue 5-HT,VIP,ICC,to explore the therapeutic mechanism of the prescription on C-IBS.Methods:Thirty-eight SPF Wistar rats,six rats were randomly selected as normal group,the remaining 32 rats were modeled by ice water for 14 days,evaluation model,30 rats were randomly selected and randomly divided into western medicine group,model control group,the Chinese medicine high,medium and low dose group,each group had 6rats.Treatment groups were respectively given Mosapride,Yun Spleen Rou Liver Recipe low,medium and high dose,model control group and blank group were given equal amount of distilled water to fill the stomach.After treatment,observe the general situation of rats.The contents of 5-HT,VIP and SP in serum were determined by Enzyme-linked immunosorbent assay.The contents of 5-HT,VIP and ICC in the colon tissue mucosa of rats were detected by immunohistochemical quantitative analysis.Results:1.General situation:After 14 days of ice water intragastric,Compared with the normal group,the fecal granules and the water content in the model control group decreased,the spirit was weak,the dorsal hair was dull,the activity was poor,and appeared the symptoms of constipation;After treatment of 14 days,compared with the model control group,treatment groups rats back hair luster,active,generally close to the normal group,the number of fecal granules and the water content increased significantly(P<0.05).2.Gastric retention rate and intestinal propulsive rate:After treatment for 2 weeks,compared with blank group,model control group gastric retention rate and intestinal propulsive rate obvious difference(P>0.05),compared with model control group,treatment groups gastric retention rate obviously reduced(P<0.05),intestinal propulsive rate increased(P<0.05).Compared with model control group,Yun Spleen Rou Liver Recipe medium dose group has significant differences(P<0.01).3.Serum:Compared with blank group,the expression of 5-HT,VIP,SP of model control group has obvious difference(P<0.05),compared with the model control group,the expression of 5-HT,VIP of treatment groups decrease(P<0.05),the expression of SP increased(P<0.05).4.Colon tissue:Compared with blank group,the positive expression of 5-HT,VIP,ICC of model control grouphas obvious difference(P<0.05),compared with the model control group,the expression of 5-HT,VIP of treatment groups decrease(P<0.05),the expression of ICC increased(P<0.05).Conclusion:1.Yun Spleen Rou Liver Recipe can obviously increase C-IBS model rats fecal grains and water content,reduce gastric residual rate and accelerate intestinal propulsive rate.2.After treatment,the expression of 5-HT,VIP of treatment groups decrease,the expression of SP,ICC of treatment groups increased.Its mechanism of action may be reduce the concentration of 5-HT and VIP in the serum and intestinal tissue,increase the SP,ICC in the intestinal tissue and Serum SP concentration to regulate the intestinal function,adjust the brain gut axis abnormal expression,which plays a therapeutic role in C-IBS.
