Gallbladder cancer is the sixth most common cancer involving gastrointestinal tract,but it is the most common malignancy of the biliary tract.This tumor is a highly lethal disease with an overall 5-year survival of less than 5% and mean survival mere than 6 months.An early diagnosis is essential as this malignancy progresses silently with a late diagnosis.At present,the early diagnosis of GBC is still more difficult,so it is a hot spot to look for more sensitive tumor markers.It is suggested that the Golgi apparatus is not only involved in the processing and transport of proteins,but also involved in the signal transduction between cells.The abnormal function of Golgi apparatus will affect the occurrence of tumor.The study of the related proteins of Golgi apparatus may provide a new way for us to study the occurrence of tumor.GP73,also known as GOLM1 and GOLPH2,is a type II Golgi transmembrane protein.The specific function of GP73 is still unclear.It is reported that GP73 is a specific protein in the cell,which exists in the normal tissues,such as stomach,prostate,colon and so on.Recent studies have shown that it was over-expressed in a variety of malignant tumors,such as HCC(hepatocellular,HCC),prostate cancer,bile duct cancer and lung cancer.These results suggest that GP73 may play a important role in the occurrence of tumor.Ki-67 is a nuclear antigen that is located in the nucleus of proliferating cells,and it has been widely used to estimate growth fraction in different cancer lesions.In this study,we detected the expression of GP73 and Ki-67 in GBC,and discussed the relationship between the expression and clinical characteristics.The relationship between the two indexes was explored.Survival analysis was used to explore the relationship between the expression of the two and the clinical prognosis of patients.Objective To investigate the expressions of Golgi protein-73(GP73)and Ki-67 antigen in GBC,and to analyze their correlation with clinical characteristics and prognosis of GBC.Methods 1.Streptavid-in-peroxidase(SP)immunohistochemistry was used to detect the expression of GP73 and Ki-67 in surgically resected specimens of 58 gallbladder carcinomas,15 gallbladder adenomas and 15 normal gallbladder samples.2.Analysed the correlationship between the three groups from the expression level of protein,and explored the relationships between their expression level and clinical pathological factors,such as the age,lymph node metastasis,Nevin staging,tumor differentiation and so on.3.Survival analysis was used to explore the relationships between the expression of GP73 and Ki-67 and the prognosis of patients.Results 1.The positive rates of GP73 and Ki-67 proteins in gallbladder carcinomas were respectively 72.4%,67.24%,significantly higher than gallbladder adenomas(GP73:40.0%,Ki-67:26.7%,P<0.05)and normal gallbladder samples(GP73:13.3%,Ki-67:20.0%,P<0.05).2.The expression of GP73 was positively correlated with tumor differentiation,Nevin staging and lymph node metastasis(P<0.05),and the expression of GP73 was positively correlated with tumor differentiation,Nevin staging(P<0.05).GP73 expression was positively correlated with Ki-67 expression in gallbladder carcinoma(r=0.473,P<0.001).3.Patients with negative expression of GP73 and Ki-67 had longer survival time than those with positive expression of GP73.Conclusion 1.Compared with the tumor adjacent tissues,the expression of GP73 and Ki-67 in gallbladder carcinoma tissues were significantly increased.The expression of GP73 and Ki-67 was associated with proliferation,invasion of gallbladder carcinoma.2.The positive expression of GP73 and Ki-67 was associated with the negative prognosis of patients of gallbladder carcinoma,combined detection of GP73 and Ki-67 is conducive to judging the progress and prognosis of the gallbladder carcinoma.
