Objective EEIHL was isolated from Inula helenium L.in our previous work.The aim of this study is to investigate the antitumor effect of EEIHL in pancreatic cancer cell line Capan-2 and CFPAC-1,to explore its underlying antitumor mechanisms,and thereby provide alternative therapeutic schedule for clinical pancreatic cancer therapy.Methods CCK8 assay and colony formation assay was used to detect the inhibitory effect of EEIHL in various pancreatic cancer cell lines.Flow cytometry was used to determine the cell cycle distribution and apoptotic cells.DAPI staining assay was used to detect the apoptotic bodies within cell nucleus.JC-1 staining was used to determine the mitochodria membrane potential.BioVision colorimetric caspase-3 assay was used to determine the activities of caspase-3 in the cell lysates.Analysis of metastasis by Wound-healing assay.Analysis of mRNA by Real-Time reverse transcription.Western blot assay was used to detect protein expression.Results EEIHL inhibited cell proliferation in a variety of solid tumor cell lines.The IC50 of EEIHL at 48h in pancreatic cancer Capan-2 and CFPAC-1 cells was 4.68 ± 0.17 μg/ml and 3.47±0.60 μg/ml.Colony formation assay also showed that EEIHL inhibited cell proliferation in a dose-dependent manner.Flow cytometry assay showed EEIHL induced G0/G1 phase arrest in pancreatic cancer cell line CFPAC-1.The enhanced apoptosis induced by EEIHL was accompanied by mitochondrial depolarization and PARP cleavage in the Capan-2 and CFPAC-1 cells.We also found EEIHL could inhibit the migration.Importantly,EEIHL could inhibit expression of snail mRNA,E-cadherin.Conclusion EEIHL induced significant apoptosis in pancreatic cancer Capan-2 and CFPAC-1 cells.EEIHL exerted synergistic anticancer efficacy against cancer Capan-2 and CFPAC-1 cells in vitro.And EEIHL inhibited the migration of the CFPAC-1 cell.
